RESUMO

The origin of precision medicine was discussed in this article,as well as its advantages and deficiencies at this stage.Tangshen formula was taken as an example to explain a rational and ideal mode of traditional Chinese medical (TCM) precision medicine research under the guidance of TCM theory.In view of the two major problems existing in the TCM modernization research,including the separated strategy among formula-disease-syndrome and independent research approach using genomics,proteomics and metabonomics,critical scientific issues on developing the drug system to biological system research mode and solutions for integrating formula-disease-syndrome were proposed here,and the roadmap and total framework of top design were also provided.The specific methods and the corresponding research results on Tangshen formula at the three stages concluded as formula-disease related,disease-syndrome combinated and formula-disease-syndrome integrated were introduced and summarized.At this time for the innovative breakthrough development of TCM,precision medicine will promote the TCM modernization research,and also will provide a powerful driving force for the establishment of holistic systems medicine.

RESUMO

Objective To study the main factors affecting the preparation of osthol ( Ost ) loaded solid lipid nanoparticies ( SLN ) . Methods The SLN were prepared by melt-homogenization method. The optimum formulation and process were selected by orthogonai design. The shape, particle size, loading capacity were investigated. Results The obtained Ost-loaded SLN were sphere or oval at a range of 100-200 nm, and were well distributed without adhesion, the loading capacity was 59. 78%. Conclusion The melt-homogenization method is available for the preparation of Ost loaded SLN.

RESUMO

Applications of network pharmacology are increasingly widespread and methods abound in the field of drug development and pharmacological research. In this study, we choose rosiglitazone compound as the object to predict the targets and to discuss the mechanism based on three kinds of prediction methods of network pharmacology. Comparison of the prediction result has identified that the three kinds of prediction methods had their own characteristics: targets and pathways predicted were not in accordance with each other. However, the calcium signaling pathway could be predicted in the three kinds of methods, which associated with diabetes and cognitive impairment caused by diabetes by bioinformatics analysis. The above conclusion indicates that the calcium signaling pathway is important in signal pathway regulation of rosiglitazone compound, which provides a clue to further explain the mechanism of the compound and also provides a reference for the selection and application of methods of network pharmacology in the actual research.

RESUMO

Major breakthrough and challenge that encountered by modern western medicine were described in this article. Some standpoints about the necessity of innovative revolution for the development of traditional Chinese medicine, together with the key points should be bring to the attention during ideological emancipation have been put forward here. Chinese scientists agreed that it is the most appropriate timing for us to establish a new medicine system, which we named Holistic Systems Medicine (HSM). The concept and research contents of Holistic Systems Medicine were also introduced in this article. Finally, a proposal for setting up“China Holistic Systems Medicine development project” was proposed.

RESUMO

In this paper, Chinese medicine theory about Fei Qi being responsible for administration and regulationwas carried out as a topic. The modern scientific research achievement had been integrated to propose a hypothesis about the function of Fei Qi that related with postganglionic fibers dominated autonomic effector's physiological functions. By analyzing the mechanism and clinical application of adrenergic receptors and cholinergic receptors agonist or antagonist drugs, and summarizing some related effective compounds from traditional Chinese medicine (TCM), we tried to clarify the basic role of Fei Qi is checking the balance of yin and yang, which is consistent with sympathetic and parasympathetic nerve regulated functions. After interpreting of modern biological knowledge, we aimed to develop a systematic integrated medicine which fused eastern and western wisdom. Finally, the guiding role of the theory derived from visceral manifestation should be expected fully in the research and development of modern drugs. The application of this theory will be a profound meaning to lead the prevention and clinical treatment for a variety of refractory diseases.

Objective To study the preparation and stability of paclitaxel self-microemulsifying drug delivery system ( PTX-SMEDDS) . Methods The formulation of the PTX-SMEDDS was optimized by monitoring the appearance, particle size, concentration, relative substance, and bacterial endotoxin. Influence of different pH and different dosage of activated carbon on PTX-SMEDDS was investigated. Subsequently it was subjected to stability studies. Results The PTX-SMEDDS was a translucent diluted-yellow solution with a mean diameter of 19. 6 nm. High speed centrifugal test indicated PTX-SMEDDS was stable. The result of the influential factor tests showed PTX-SMEDDS was unstable to high temperature and light. In the accelerated tests for 6 months, the quality of PTX-SMEDDS was stable. The samples should be sealed and stored at low temperature and shady place. Conclusion Easily prepared and stable formulation of PTX-SMEDDS is achieved, and further investigation is warranted to develop self-microemulsifying drug delivery system for injection.

RESUMO

The objective of this study is to fully investigate the therapeutic effect and mechanisms of action of Gegen Qinlian decoction (GD) on type 2 diabetes mellitus (type 2 DM). A rat model of type 2 DM was established with the combination of high-fat diet and multiple low doses of streptozotocin (STZ). Biochemical indicators related to glucose metabolism disorders, insulin resistance, oxidative stress were observed. The type 2 DM rats were administrated with GD for 80 days, the above-mentioned indexes were detected. The results indicated that the hepatic glycogen synthesis level was promoted, fasting blood glucose level and fasting blood insulin level were significantly reduced, insulin sensitivity index was significantly improved; the level of superoxide dismutase (SOD) was increased and the level of malondialdehyde (MDA) was reduced; pathologic morphology of pancreas and kidney was ameliorated in the GD group. It was indicated that the therapeutic mechanisms of action of GD on type 2 DM might be related to its effect of ameliorating glucose metabolism disorders, relieving insulin resistance, increasing the tissues' sensitivity to insulin, improving the antioxidative ability of living system, GD has therapeutic effect on type 2 DM and protective effects against damaged pancreatic function.

This study was aimed to screen antibacterial agents against 9 pathogenic bacteria from 240 extracts of 60 traditional Chinese medicinals ( TCM ) . And the minimum inhibitory concentration ( MIC ) test was applied on extracts with positive results. The disk diffusion was employed to screen the antibacterial activity preliminar-ily among 240 extracts. The MICs of active extracts were tested by liquid culture method (double dilution method). The results revealed that 104 extracts show antibacterial activity on one or more strains, 20 of them show strong inhibition on three commonly seen bacteria ( MIC < 0 . 2 mg/mL ) . It was concluded that ethanol , acetone and hexane extracts of 11 TCMs including Salvia miltiorrhiza, Cnidium monnieri, Polygonum tinctorium, Taraxacum mongolicum, Morus alba, Glycyrrhiza uralensis, Curcuma longa, Arnebia euchroma, Lobelia chinen-sis , Chrysanthemum indicum and Buddleja officinalis show strong inhibition on Bacillus subtilis , Bacillus cereus , Staphylococcus aureus .

Chloroplast genome sequences have comprehensive application prospects in DNA barcoding and chloroplast engineering in traditional Chinese medicine. The complete chloroplast genome of Magnolia officinalis sequenced by high-throughput pyrosequencing and a sequencing procedure was established. Fourteen contigs were obtained after de nove assembly. The sequencing percent of coverage was 99.99%. The chloroplast genome is 160 183 bp in size, and has a typical quadripartite structure with the large (LSC, 88 210 bp) and small copy (SSC, 18 843 bp) regions separated by two copies of an inverted repeat (IRs, 26 565 bp each). chloroplast genes were successfully annotated, of which 17 genes located in each IR region. The chloroplast genome features in Magnolia officinalis are nearly identical to those from other Magnoliid chloroplast genomes. Phylogenetic analyses were performed based on 81 shared coding-genes for a total of 9 Magnolia samples of 5 closely related species. Results showed that distinguishing among species was generally straightforward at the species and population level. This study confirmed the effectiveness of our chloroplast genome sequencing procedure. The chloroplast genome can provide distinguishing differences to help identify Magnolia officinalis and its closely related plants.

<p><b>OBJECTIVE</b>To verify the rational of Chinese medicine Shuanglong formula.</p><p><b>METHOD</b>Rat models of acute myocardial ischemia were induced by hydrochloride isoproterenol. ECG J point change, myocardial infarction area and cellular enzyme (CK, LDH, SOD and MDA) levels were observed and detected to show protective effect of treatment groups.</p><p><b>RESULT</b>The single drug prescriptions and compatibilities both could improve ECG performance, decrease levels of CK, LDH, SOD and MDA in serum, and reduce a certain myocardial infarct size.</p><p><b>CONCLUSION</b>The Shuanglong formula at the high dose of 3:7 was proved to be more effective on myocardial ischemia.</p>

<p><b>OBJECTIVE</b>To establish a full-quantified fingerprint method for analyzing Chinese herbal additives by HPLC.</p><p><b>METHOD</b>A HPLC in combination with PDA detector was applied with a phenomenex luna C18 (4.6 mm x 250 mm, 5 microm) column by gradient elution using acetonitrile-0.1% formic acid solution as the mobile phase. The flow rate was 1 mL x min(-1) and the detection wavelength was set at full spectrum scan.</p><p><b>RESULT</b>According to the selected chromatographic conditions, the full-quantified fingerprint of the Chinese herbal additive has good precision, reproducibility, stability and recovery.</p><p><b>CONCLUSION</b>The HPLC method developed for simultaneous determination of seven compounds is simple and valid. It can be used for quality evaluation and quality control of Chinese herbal additive and its processing products.</p>

RESUMO

This study was to report the effect of Tangshen Formula on phospholipids metabolism in diabetic nephropathy patients. A normal phase-HPLC-TOF/MS method was used in this study for the determination of seven species of phospholipids in human plasma. Then, the concentration changes of potential phospholipids biomarkers were discussed in diabetic nephropathy phase III and phase IV patients among different groups, including before and 3, 6 months after administration of Tangshen Formula. Significant increases of PE750, PI885, PC792, PC826, PC830, PC854 and PC802 levels were observed 6 months after administration of Tangshen Formula and conventional western medicine, as well as a decrease of LPC540 level, when compared with those before medication. Concentrations of all the potential phospholipids biomarkers showed a tendency towards normal levels; however, both the improvement degree and onset time of these compounds were not same. Additionally, Tangshen Formula treatment based on conventional western medicine treatment was more efficient in adjusting the levels of these compounds when compared with western medicine treatment alone, especially for the phase IV patients. These results indicated that Tangshen Formula was capable in regulating and improving phospholipids metabolism in diabetic nephropathy patients, which may be related with the direct or indirect inhibition of protein kinase C pathway and the corresponding reduction of phospholipase A2 activity. Therefore, Tangshen Formula may be used as an effective drug for diabetic nephropathy therapy, at least as an adjunctive therapeutic drug.

<p><b>OBJECTIVE</b>To establish the method for the quality control of Xuefuzhuyu granule.</p><p><b>METHOD</b>The content of Semen Armeniacae Amarum PE, Hydroxysafflor yellow A, paeoniflorin and ferulic acid in Xuefuzhuyu granule were determined by HPLC under multi-wavelength of 210, 403, 230, 316 nm.</p><p><b>RESULT</b>The calibration curve of Semen Armeniacae Amarum PE. hydroxysafflor yellow A, paeoniflorin and ferulic acid were linear within 0.1154-0.9232 (r = 0.9998), 0.0219-0.1754 (r = 0.9994), 0.3952-3.1616 (r = 0.9995), and 0.0426-0.3408 microg (r = 0.9998), respectively. The average recoveries were 101.78% (RSD was 1.7%), 99.60% (RSD was 2.9%), 98.90% (RSD was 2.0%), and 100.31% (RSD was 1.8%), respectively.</p><p><b>CONCLUSION</b>The method of quantification is accurate, rapid and reliable for the quality control of Xuefuzhuyu granule.</p>

<p><b>OBJECTIVE</b>To establish a HPLC method for the determination of 11 essential compounds in Xuebijing injection.</p><p><b>METHOD</b>Chromatogaphic analysis was performed on Aglient Zorbax SB-C18 (4.6 mm x 250 mm, 5 microm). The mobile phase 0. 5% acetic acid and methanol-acetonitrile-acetic acid (40: 60: 0.5). The flow rate was 1.0 mL x min(-1) with the detection wavelength at 280 nm; column temperature at 35 degrees C, and automatic sampling volume of 20 microL.</p><p><b>RESULT</b>All the 11 essential compounds showed good linearity (r = 0.9982-0.9999)in the range of the tests concentration. The RSD of the precision, reproducibility and stability tests were less than 3%; the average recoveries of the method were in the range of 95.02%-104.94%.</p><p><b>CONCLUSION</b>The method is simple, rapid and accurate, so it is proposed for the quality control of compounds of Xuebijing injection.</p>

<p><b>OBJECTIVE</b>To establish a method to evaluate the in vitro release of Chinese medicinal compound sustained release preparations using multi-index chromatographic fingerprint.</p><p><b>METHOD</b>With the Chinese medicinal compound Jiangya sustained tablets as model preparation, the in vitro cumulative release of the main component flavones and fingerprint were determined by HPLC fingerprint. Aacacetin, rutin, luteolin, fingerprint were determined simultaneously and used to calculate the in vitro cumulative release of the tablets.</p><p><b>RESULT</b>Aacacetin, rutin, luteolin, and the fingerprint had different cumulative release.</p><p><b>CONCLUSION</b>Multi-index chromatographic fingerprint can reflect different content of multi components of preparations and can be used in the evaluation on in vitro release of the sustained release preparations of Chinese medicinal compound. The method is simple, rapid, and of multi information.</p>

RESUMO

Objective To study the molecular mechanism of acute myocardial infarction(AMI),searching for protective genes and injury-related ones,aimed to fight myocardial infarction disease.Methods Rat model of AMI was established with left anterior descending (LAD) coronary artery ligation.Gene chip was used to verify the mechanism of AMI at molecular level.Results The author screened out 190 differential expression genes between AMI rat myocardium and sham operated rat myocardium in the gene expression profile study;In the Model/Sham operation gene expression profile,there were 89 genes depressed expression and 101 genes elevated expression.Most of them involve in the 12 known categories,and relate to the metoblolism,signal and trandport pathways.Conclusion The major possible mechanism of AMI is to regulate the local expression of related gene through a multi-pathway.

This paper reviews recent development and applications of high-speed countercurrent chromatography (HSCCC) in the separations on the analytical scale,the semi-preparative and preparative scale of natural products,antibiotics,proteins,inorganic compounds etc.Several series of two-phase solvent systems to facilitate the systematic search suitable for HSCCC are introduced.And the new countercurrent chromatography (CCC)technologies such as HSCCC coupled with MS,pH-zone-refining countercurrent chromatography,and ion-pairing Countercurrent Chromatography are explained and prospected in term of applications.

By comparing with the molecule weights, retention times and product ion spectra of the standards, the cholic acid, deoxycholic acid and chenodeoxycholic acid were identified in the traditional Chinese compound medicine——“Qingkailing” injection by high performance liquid chromatography/mass spectrometry/mass spectrometry. Two other isomers of cholic acid and deoxycholic acid were also found, but tieir detail structure was still unknown due to the absence of standards. A quantitative method to determine cholic acid in the medicine was established and studied. An internal standard was used in quantitative analysis. When the concentration of cholic acid is from 875 ng/L to 140 μg/L, the chromatographic peak area and concentration are of good linear relation. The linear correlation coefficient was determined to be 0.9999 and the RSD is 2.04%. This method is simple, selective and highly sensitive, which is helpful for the quality control of traditional Chinese medicine.

0.999). The relative standard deviation (RSD) of chromatogram area was less than 1.0% for all of them after six successive injections. The detection limit was 0.05 ?g/mL for norephedrine (NE), 0.04 ?g/mL for norpseudoephedrine (NPE), 0.1 ?g/mL for E and PE, and 0.2 ?g/mL for mephedrine (ME) and mepseudoephedrine (MPE), respectively (S/N≥3). The recovery rate was more than 97.1% for six ephedrine alkaloids. Under this method system, all of the above-mentioned six samples were tested and found to contain some of the impurity at different levels. Conclusion This developed method, which is very simple, perfect precision, high sensitivity, and selectivity, can be used for the qualitative and quantitative determination of impurities of ephedrine alkaloid-type samples.

Objective To study the reaction mechanism of ginsenosides in red ginseng processing. Methods The major constituents of ginsenosides were assigned by HPLC/MS/MS data. Ginsenosides (including protopanaxadiol, protopanaxatriol, and oleanolic acid type) of white ginseng and red ginseng were compared by HPLC/MS data. Results Ginsenosides were produced from esterolysis of corresponding malonyl ginsenosides in red ginseng processing. Another reaction of malonyl ginsenosides in red ginseng processing was decarboxylated to produce corresponding acetyl ginsenosides. Hydrolysis of glycosidic bond at C_(20) and isomerous reaction of C_(20) were the main reaction of dammarane type ginsenoside. High degree of esterolysis and glycosidic bond hydrolysis of oleanolic acid type ginsenosides all occurred during the red ginseng processing. Conclusion Great changes of ginsenosides have been obviously found in processing ginseng to red ginseng. Special constituents of red ginseng could be produced in ginseng processing.
