RESUMO

Objective:To investigate interleukin (IL)-36 expression in patients with myasthenia gravis (MG), and to study the modulatory function of IL-36 on regulatory T cells (Tregs) and Th17 cells in MG patients.Methods:Fifty-one MG patients (MG group) and 25 healthy controls (control group) were enrolled in this study in Xinxiang Central Hospital between July 2016 and August 2021. Peripheral blood was collected. Plasma and peripheral blood mononuclear cells (PBMCs) were isolated. Plasma IL-36α, IL-36β, IL-36γ, IL-36RA, IL-35, and IL-17 levels were measured by enzyme-linked immunosorbent assay. The percentages of Tregs and Th17 cells were measured by flow cytometry. Forkhead box protein P3 (FoxP3) and retinoid-related orphan receptor gamma t (RORγt) mRNA expressions were measured by real-time polymerase chain reaction. PBMCs or purified Tregs from MG patients were stimulated with recombinant IL-36β (5 ng/ml). Changes of Tregs and Th17 cell percentages, IL-35 and IL-17 secretions, FoxP3 and RORγt mRNA expressions, as well as immunosuppressive activity of Tregs were analyzed.Results:There were no statistically significant differences of IL-36α, IL-36γ, or IL-36RA between the control group and the MG group (all P>0.05). IL-36β level was notably higher in the MG group compared with the control group [(73.43±13.91) pg/ml vs (60.91±12.65) pg/ml, t=3.79, P<0.001]. Treg percentage [(4.67±1.33)% vs (6.32±1.81)%, t=4.48, P<0.001], IL-35 [(50.06±7.93) pg/ml vs (65.37±8.90) pg/ml, t=7.59, P<0.001] and FoxP3 mRNA expression (1.03±0.14 vs 1.57±0.46, t=7.78, P<0.001) was lower, while Th17 cell percentage [(1.05±0.15)% vs (0.94±0.21)%, t=2.61, P=0.011], IL-17 [(40.61±13.13) pg/ml vs (33.09±11.48) pg/ml, t=2.44, P=0.017] and RORγt mRNA expression (1.26±0.16 vs 1.03±0.13, t=6.08, P<0.001) was higher in the MG group ( P<0.05). There were no statistically significant differences of above indices between different genders, onset ages, afflicting with thymoma, or different Osserman types (all P>0.05). There were no statistically significant correlations between above indices and quantitative myasthenia gravis (QMG) score (all P>0.05). Recombinant IL-36β stimulation did not affect PBMCs proliferation in MG patients ( P=0.248), and reduced Tregs percentage [(3.05±0.66)% vs (4.18±1.07)%, t=4.23, P<0.001], IL-35 secretion [(48.12±10.93) pg/ml vs (56.96±13.73) pg/ml, t=2.36, P=0.023] and FoxP3 mRNA expression (0.99±0.17 vs 1.18±0.13, t=4.01, P<0.001), but did not affect Th17 cell percentage, IL-17 secretion or RORγt mRNA expression (all P>0.05). Recombinant IL-36β stimulation inhibited immunosuppressive activity of Tregs, which presented as enhanced cellular proliferation [(0.83±0.12)×10 5vs (0.69±0.15)×10 5, t=3.02, P=0.005] and reduced IL-35 secretion [(28.71±10.08) pg/ml vs (37.12±10.47) pg/ml, t=2.39, P=0.023]. Conclusion:Increased IL-36β contributed to the regulation of Tregs/Th17 cell balance probably through inhibition of Tregs function in MG patients.

RESUMO

The JAK-STAT signaling pathway, composed of Janus kinase (JAK) and signal transducer and activator of transcription (STAT), is an important intracellular signaling pathway. It is involved in the occurrence of many inflammatory diseases. Blocking this pathway by JAK inhibitors has become an effective approach to the treatment of some diseases, such as rheumatoid arthritis and ulcerative colitis. However, JAK inhibitors have not been approved for the treatment of skin diseases at present. This review summarizes application prospects of JAK inhibitors in the treatment of skin diseases.

RESUMO

The homeobox(HOX)gene is a special type of transcriptional regulator that plays an important role in physiologi-cal regulation mechanisms such as embryonic development,cell growth,differentiation and migration.Recent studies have found that HOX gene expression is abnormal and affects the occurrence and development of tumor.This review summarizes the progress of HOX gene and gynecologic oncology including cervical cancer,endometrial cancer and ovarian cancer,and the instruction and application of HOX gene in the treatment of cancer.We are making the guiding direction of exploring HOX gene through summarizing the existed re-search reports in order to alleviate the pain of gynecological cancer patients.

OBJECTIVE:To establish the method for rapid determination of nuezhenoside in Salvia miltiorrhiza.METHODS:The contents of salvianolic acid B,rosmarinic acid,lithospermic acid and tanshinone (tanshinone Ⅱ A+tanshinone Ⅰ +cryptotanshinone) were determined by HPLC (as reference value).Quantitative model for the contents of above components was established by partial least square (PLS)-NIR spectra.According to the reference value,143 samples were collected and the spectrum was pretreated by first-order derivative.The optimal range of wave band for salvianolic acid B,rosmarinic acid,oxalic acid and tanshinone were 6 773.98-3 981.12,6 670.85-3 996.54,8 544.66-3 936.28,8 188.06-3 875.31 cm-1.RESULTS:The methodology for the content determination of salvianolic acid B,rosmarinic acid,oxalic acid and tanshinone were in line with the requirement.The coefficient (R2) of internal cross validation for quantitative correction model of salvianolic acid B,rosmarinic acid,oxalic acid and tanshinone were 0.919 0,0.832 2,0.821 5,0.925 6.The deviation of corrected mean square roots were 4.46,0.48,1.34,0.71,respectively.The R2 values of external validation were 0.852 6,0.957 3,0.819 3,0.953 1,respectively;and mean square root of prediction error (RMSEP)were 9.77,0.28,0.94,0.63,respectively.CONCLUSIONS:The method is rapid,accurate,simple and pollution-free,and can be used for rapid determination of multi-maker ingredients in S.miltiorrhiza.

Objective:To optimize the formula of 5-aminosalicylic acid enema in situ gel. Methods:5-Aminosalicylic acid ene-mas in situ gel was prepared using a cold dissolution method with carbomer as the gel matrix and xanthan gum as the thickener. A 32 full factorial design was used to investigate the effects of the concentrations of carbomer and xanthan gum on the viscosity before and af-ter the gelling, duration of inversion tube and sedimentation rate. Response surface methodology was used to optimize the formula. Re-sults:The quantitative relationships between the two factors and the four evaluation indices were obtained. The optimum formula was as follows:the concentration of carbopol and xanthan gum in the enema was 0. 7% and 0. 15%, respectively. The viscosity before and af-ter the gelling was 500-1 000 mPa·s and 2 200-2 700 mPa·s, respectively. The duration of inversion tube test was 40-80 min and the sedimentation rate was more than 98. 5%. Conclusion:The multi-objective simultaneous optimization of the formula of 5-aminosal-icylic acid enema in situ gel is accomplished by factorial design and response surface methodology.

Objective:To establish an HPLC method to determine 5-amino salicylic acid (5-ASA) and its related substances in the enemas. Methods:The liquid chromatograph was equipped with a Hypersil ODS C18 column (250 mm × 4. 6 mm,5. 0 μm) with gradient elution. The mobile phase A was composed of pH 7. 5 phosphate buffer ( PBS), tetrabutyl ammonium hydroxide solution (TBAH), methanol and water (600∶50∶50∶300), the mobile phase B was composed of pH 7. 5 PBS, TBAH, methanol and water (200∶50∶300∶450). The wavelength was 208nm, the column temperature was 40 ℃ and the injection volume was 20μl. Results:5-ASA and its two degradation products could be completely separated. The concentration of 5-ASA and the corresponding peak area had a good linear relationship within the range of 1-250μg·ml-1(r=0. 999 5). The average recovery was 99. 44%(RSD=0. 94%,n=9). Conclusion:The method is simple, sensitive and reproducible, and can be used in the determination of the content and related substances in the enemas.

RESUMO

To develop standard in vitro chondrosarcoma models, we synthesized three hydrogels (i. e., PDMAAm, PNaAMPS and PMETAC) and investigated the influence of Young's modulus, swelling ratio and electric charges on the behavior of chondrosarcoma cells seeded on the hydrogels, including morphology, adhesion and aggregation. Results showed that the morphology of chondrosarcoma cells at 6h was dependent on the charges of hydrogels; cells present spindle-shaped and round-shaped morphology on negative charged and neutral hydrogel, respectively, while no cells spreaded on positive charged hydrogel. Chondrosarcoma cells formed aggregates on neutral PDMAAm after further culture. The hydrogels can be synthesized easily and has the characteristics of ease at use with defined components, which holds great potential for developing standard chondrosarcoma models in vitro.

RESUMO

Objective To explore the feasibility and validity of Satir model on compulsory drug rehabilitation for female drug users in closed settings. Methods Satir Model-based group psychotherapy was adopted in the treatment on 33 female drug users. Results The findings are that the pre-treatment SCL-90 scores were all higher than norm,indicating very high significant differences(P<0.01) ; while the post-treatment SCL-90 scores,except the score of somatization, were approximate to norm, showing no statistical differences(P >0. 05). Conclusion Satir model played a positive role in the process of female drug users mental health recovery.

RESUMO

HIF-1α, a transcriptional activator regulating cells and tissues' response to hypoxia, can induce expression of a variety of factors and facilitate adaptation to hypoxia. Tumor hypoxic microenvironment and expression of HIF-1α is closely related to the occurrence of colorectal cancer, development, invasion, metastasis and prognosis, which becomes a hot spot in colorectal cancer research. To further clarify the process of colorectal cancer and to explore new treatment methods in colorectal cancer, HIF-1α in colorectal cancer studies was briefly reviewed.

Homeodomain-interacting protein kinase 2(HIPK2)is a member of a member of the serine/threonine protein kinase family which localized in the mucleus.It is not only involved in advanced stage embryogenesis,development of nerve tissue,retina and muscular tissue,but also takes part in regulation of tumor signaling transduction,down-modulated expression of oncogene,induced apoptosis of tumor cells,and inhibited angiogeuesis of tumor.The regulation of HIPK2 in tumor signaling transduction pathway is associated with P53 signaling,Wnt/β-catenin pathway and hypoxia.

Objective To establish cell models with different insulin sensitivity status and examine the expression of protein kinase B (PKB/Akt) in the same conditions. Methods 3T3-L1cells were cultured with different glucose concentrations (3.0, 5.5 and 50.0 mmol/L respectively).Insulin sensitivity, as presented by insulin-induced glucose transport rate in the cells, was tested by 3H-2-DG incorporation. PKB expression in cells was determined by RT-PCR. Results The insulin-induced glucose transport rate in 3T3-L1 cells was affected by different concentrations of glucose. The transport rate of 3H -2-DG in cells cultured in low glucose concentration was increased by 152% and 50% compared with high glucose concentration and control group, respectively. There was no difference in PKB mRNA expression among the three groups under the same conditions,whereas there was an obvious reduction of PKB expression in the three groups after the cells were treated with glucose and insulin. Conclusions 3T3-L1 cells cultured with different concentrations of glucose present the different insulin sensitivity status, but the difference of PKB mRNA expression has not been observed in the same condition.

BACKGROUND: Ultra-high molecular weight polyethylene (UHMWPE) is widely used as acetabulum. The hip joint will produce lots of grind particles and lead to loosening of artificial joint because of alternating load and bad lubricate condition.OBJECTIVE: To learn about the lubricate mechanism of joint, introduce the present situation on artificial hip joint of UHMWPE including wear behavior and wear resistance properties, and grope for scientific evaluate method of tribological properties in artificial joints.RETRIEVAL STRATEGY: The articles published between January 2000 and October 2007 were searched by using computer with the key terms "hip joint, ultra-high molecular weight polyethylene, tribology" in CNKI, Weipu Science and Technology Database, Wanfang Database, and its language was limited to Chinese. Meanwhile, the English articles were searched in IEEE/IEE Electronic Library and Elsevier Electronic Journal Database with the key terms "hip simulators, ultra-high molecular weight polyethylene, tribology ". Those referring to the friction and wear property of the joints were included, whereas repeated researches were deleted.LITERATURE EVALUATION: Totally 729 articles were received. After the primary screening of the titles and abstracts, 601 repeated or similar researches were excluded, and other 128 articles were adopted for further analysis, finally 22 articles in accordance with the inclusion criteria were included as references.DATA SYNTHESIS: The study about UHMWPE focused two points, one is surface modification, the other is wear and friction mechanism.①The study indicates that wear properties of UHMWPE can be improved by surface modification, such as ion implantation and nanometer padding have a remarkable elevating ability of the wear and mechanical properties.②At recent years, scholars do many basal experiences about the tribology of hip joints, and have approved that wear rate of friction pairs would reduce under biological lubricate condition using artificial joint simulators or traditional tester in various conditions.CONCLUSION: The study about hip joint in China drops behind the developed counties, this traditional method can not really reflect wear and friction properties in vivo. Therefore, the following research should include making clear the relationships of load, environmental condition and wear of artificial joint in vivo, as well as groping for a scientific evaluate method of tribological properties in hip joints.

@# ObjectiveTo observe the effect of rehabilitation treatment on the functional recovery in stroke patients with hemiplegia. Methods31 stroke patients with hemiplegia receiving rehabilitation treatment and physical therapy for one to two months were classified as treatment group, and at the same time another 30 stroke inpatients with hemiplegia were chosen for control group without receiving rehabilitation treatment. Two groups were compared to each other according to Fugl Meyer functional scores admision and discharge. ResultsFunctional scores of the two groups at discharge were higher than those at admision. Functional scores in the rehabilitation group were markedly higher than those in the control group(P<0.01). ConclusionsThe rehabilitation treatment has the significant effect on the functional recovery in stroke patients with hemiplegia.

ive] Therapeutic effect of western medicine combined with Chinese herbal medicine for central serous chorioretinopathy (CSC) is observed. [Methods] Sixty-four patients with CSC were ran domly allocated to Group A ( n = 33) and Group B ( n = 31) . Group B were treated only with routine western medicine, and Group A added with Chinese herbal medicine with the actions of nourishing liver and kidney, invigorating spleen for eliminating dampness and activating blood-flow to remove blood-sta sis. Plasma catecholamine (CA) content and the activities of tissue plasminogen activator (tPA) and plasminogen activator inhibitor (PAI) were measured before and after treatment. Objective To discuss the mechanism of large dose methylprednisolone(MP) on cerebral protection of rats after focal ischemic reperfusion.Methods Using the Wistar rat models of focal cerebral ischemic reperfusion,we observed the effects of large dose MP before and after ischemia on the volume of middle cerebral after infarction and the change of cerebral water content,meanwhile the pathological change of brain tissue was observed.Results Compared with the control group, the volume of middle cerebral after infarction in MD treatment group was remarkably reduced before and after ischemia (P 0 05 ). From the control group but not the MP treatment group, we found cerebral infiltration of macrophages from the histopathology of rat brain tissues.Conclusion Large dose MP might improve ischemic brain injury,the mechanism related to reducing volume of cerebral infarction and inhibited the infiltration of macrophages in the rat brain tissues.

Objective To investigate the relationship between transient ischemic attack(TIA) and intracranial or extracranial artery stenosis.Methods Digital subtraction angiography (DSA) was further used in 56 cases with cerebral artery stenosis from 68 TIA patients after examinations of B ultrasound of neck, transcranial color doppler(TCD) and cardiac ultrasound. Results 6 cases were shown stenosis or occlusion in middle cerebral arteries(MCA), 21 cases were shown stenosis or occlusion in arteria carotis interna,8 cases were shown stenosis in subclavian arteries and 28 cases were shown stenosis or occlusion in vertebral arteries or basal arteries. 44 cases with TIA were related to stenosis of large arteties, 16 cases of which were intracarotid TIA, and 28 cases were vertebrobasilar TIA; 18 cases were embotic TIA, 15 cases of which were intracarotid TIA, and 3 cases were vertebrobasilar TIA. 4 casea were lacunar TIA; and 2 cases were related to vascular spasm. Conclusions Most patients with TIA have cerebral artery stenosis.There is a positive correlation between TIA and intracranial or extracranial artery stenosis.

Objective To investigate the correlation between non-lacunar infarcts in the white matter of brain and intracranial or extracranial vessels stenosis.Methods Brain magnetic resonance imaging(MRI) and digital subtraction angiography(DSA) were performed in 30 patients with non-lacunar infarcts in the white matter of brain and the results were analyzed.Results Of 12 cases with focal non-lacunar infarcts in the area of basal ganglion,9 cases were shown occlusion or highly stenosis of proximal segments of unilateral internal carotid artery(ICA) in DSA,2 cases were shown highly stenosis of M1 segment of middle cerebral artery(MCA),and 1 case was normal.Of 6 cases with focal non-lacunar infarcts in the area of basal ganglion and lateral periventricular white matter,3 cases were shown occlusion or highly stenosis of proximal segments of unilateral ICA in DSA,1 case was shown occlusion of C5 segment of unilateral ICA,and 2 cases were shown occlusion of M1 segments of MCA.Of 4 cases with focal non-lacunar infarcts in periventricular white matter or centrum ovale,1 case was shown occlusion of C5 segment of unilateral ICA,2 cases were shown highly stenosis of M1 segments of MCA,and the other one was normal.Of 8 cases with subcortical superior border infarction(SSBI) or subcortical lateral border infarction(SLBI),6 cases were shown occlusion or highly stenosis of proximal segments of unilateral ICA,1 case was shown occlusion of proximal segments of bilateral ICA,and 1 case was shown occlusion of M1 segment of MCA.Conclusion There are strong correlations between focal non-lacunar infarcts in the areas of basal ganglion or lateral periventricular white matter and the stenosis or occlusion of large arteries of ICA system.
