RESUMO

<p><b>OBJECTIVE</b>To evaluate the effect of bleomyin A5 combined with phosphorus-32 colloid in the treatment of mucocele.</p><p><b>METHODS</b>A total of 214 patients divided into three groups, bleomyin A5 (50 cases), phosphorus-32 colloid (50 cases) and bleomyin A5 combined with phosphorus-32 colloid (114 cases).</p><p><b>RESULTS</b>The efficacy of bleomyin A5 group, phosphorus-32 colloid group, and bleomyin A5 combined with phosphorus-32 colloid group was 84% (42/50), 82% (41/50) and 98% (112/114), respectively. There were significant difference in efficacy among the three groups (P < 0.05). The phosphorus-32 colloid group and the bleomyin A5 group had no significant difference in efficacy (P > 0.05).</p><p><b>CONCLUSIONS</b>The independent use of bleomyin A5 and phosphorus-32 colloid is effective, but the combined use of the two methods is more effective.</p>

RESUMO

In this study, Quantitative 32P bremsstrahlung planar and SPECT imaging and consequent dose assessment were carried out as a comprehensive phantom study to define an appropriate method for accurate Dosimetry in clinical practice. CT, planar and SPECT bremsstrahlung images of Jaszczak phantom containing a known activity of 32P were acquired. In addition, Phantom contour was determined for attenuation correction and image registration. Reconstructed SPECT slices were corrected for attenuation effect using two different methods: conventional Chang's method and an expectation maximization algorithm followed by CT and SPECT image registration. Cumulated activity was calculated by a predefined calibration factor. Both attenuation correction algorithms were quantitatively assessed by the Monte Carlo SIMIND program. Acquired planar Bremsstrahlung images were quantified by the Conjugate View Method, as well. Calculated activities were statistically different among various quantification methods [P= 0.0001]. When iterative expectation maximization algorithm and applied methods were used, mean calculated activity had the least difference with real activity of +/- 3%. Quantitative 32P Bremsstrahlung SPECT imaging could accurately determine administered activity and assess radiation dose if precise attenuation correction and appropriate registration with CT were done even without sophisticated scatter correction or when SPECT/CT machines are not available. Therefore, it has the potential of specific tumor/organ dosimetry in clinical practice. The best method for calculating activity is quantitative SPECT using iterative expectation maximization algorithm. Additionally, applied method for determining phantom contour was practical for attenuation correction and image registration

<p><b>OBJECTIVE</b>To investigate the efficacy of CO2 laser combined with 32P-patch contact brachyradiotherapy for the treatment of keloids.</p><p><b>METHODS</b>From 2001 to 2006, 121 cases with 151 keloids, which reoccurred after treatment with more than 2 methods, underwent continuous CO2 laser treatment to remove the hypertrophic scar tissue, following by ultra-pulse CO2 laser to treat the fresh granulation tissue. After wound healing, 32P-patch contact brachyradiotherapy was used for the lesion, 0.5-1 MBQ/cm2 for 72-96 hours, every 1-2 months. 2-3 treatment were applied.</p><p><b>RESULTS</b>Among the 151 keloids, good result was achieved in 111 keloids, and effective result in 40 keloids. Adverse effect included hyperpigmentation in 21 lesions and hypopigmentation in 32 lesions. The patients were followed up for 2-6 years without relapse.</p><p><b>CONCLUSION</b>CO2 laser combined with 32P-patch contact brachyradiotherapy is an effective and safe method for the treatment of recalcitrant keloids.</p>

<p><b>OBJECTIVE</b>To investigate the use of a 32P application device (AD) in the treatment of condyloma acuminatum (CA) in the rectum, and to compare its clinical effect with that of the microwave therapy.</p><p><b>METHODS</b>This study included 107 cases of CA in the rectum, 99 males and 8 females, aged 21-58 (33.6 +/- 9.4) years. Forty-six of the patients (the AD group) were treated with a self-made 32P application device, which, as a tube-shaped carrier of radionuclide 32P colloid, was fixed in the rectum at the diseased part for medication at 4.9-8.2 Gy for 3-5 hours once and 1-2 times a week. The other 61 (the microwave group) were treated by microwave burning under local anesthesia. Both groups of patients were followed up for over 3 months for comparison of the therapeutic results and observation of the stability and reliability of the 32P application device.</p><p><b>RESULTS</b>The rates of cure, reoccurrence and adverse reaction were 84, 8%, 13.0% and 8.7% in the AD group, compared with 40.3%, 55.7% and 75.4% in the microwave group, with statistically significant differences between the two groups (P < 0.01).</p><p><b>CONCLUSION</b>The 32P application device, with its advantages of low cost, easy operation, good effect, high safety and reliability, low recurrence, fewer adverse events and good acceptability, is highly valuable for the treatment of CA in the rectum.</p>

<p><b>OBJECTIVE</b>To study the effects of implantation brachytherapy with delayed-release particles of 32P-chromic phosphate-poly (L-lactide) (32P-CP-PLLA) on prostate cancer (PCa) in nude mice.</p><p><b>METHODS</b>We established a subcutaneous transplantable PCa model in nude mice, and randomly divided them into six groups, Groups A, B and C implanted intratumorally with 32P-CP-PLLA delayed-release particles at 3.7, 7.4 and 14.8 MBq, Groups D, E and F with 125I particles at the same doses as the former three, and another six nude mice were included in Group G as the blank control. Then we killed the mice at 21 days after the treatment, observed the effects of the particles on the morphology of the tumor and their inhibition of tumor growth, counted WBCs and platelets (PLTs) in the peripheral blood, and detected the toxic reaction of the blood.</p><p><b>RESULTS</b>At 21 days after the treatment, the solid tumor tissues exhibited bleeding and necrotic changes, and the rates of tumor inhibition were positively correlated with the doses of administration. Groups A, B and C showed statistically significant differences from Groups D, E, F and G in the rate of tumor inhibition ([ 65.72 +/- 6.95]%, [77.58 +/- 4.32]% and [82.64 +/- 4.03]% versus [35.61 +/- 5.61]%, [43.30 +/- 6.94]% and [69.01 +/- 4.98]%), WBC count ([1.72 +/- 0.37] x 10(9)/L, [1.23 +/- 0.27] x 10(9)/L and [0.86 +/- 0.25] x 10(9)/L versus [1.45 +/- 0.40] x 10(9)/L, [0.51 +/- 0.24] x 10(9)/L, [0.37 +/- 0.26] x 10(9)/L and [3.96 +/- 0.26] x 10(9)/L), PLT count ([1.18 +/- 0.11] x 10(11)/L, [0.97 +/- 0.10] x 10(11)/L and [0.72 +/- 0.11] x 10(11)/L versus [0.97 +/- 0.15] x 10(11)/L, [0.76 +/- 0.16] x 10(11)/L, [0.64 +/- 0.12] x 10(11)/L and [2.89 +/- 0.21] x 10(11)/L) and body weight ([18.60 +/- 0.66] g, [17.60 +/- 0.39] g and [16.90 +/- 0.68] g versus [17.86 +/- 0.60] g, [15.56 +/- 0.39] g, [14.61 +/- 0.65] g and [19.95 +/- 0.73] g) (P < 0.01).</p><p><b>CONCLUSION</b>Intratumoral implantation of 32P-CP-PL-LA is a safe, simple and effective radionuclide interventional therapy for prostate cancer.</p>

RESUMO

Keloids are the result of excessive fibroblast proliferation and then over-abundant collagen deposition. There is no method able to guarantee absolute success in the therapeutic approach to keloids. Our case report involves a female patient with six lesions treated with a 32P-patch brachyradiotherapy. Pre-treatment and adjuvant treatment of the lesions were performed with thiomucase, 5-fluoruracil, procaine and triamcinolone. Taking into account the activity contained in each of the patches and the total radiation dose to be administered according to clinical practice, dosimetric calculations were done for each lesion. Separate silicone patches with chromic [32P]phosphate were designed for each lesion based on these calculations. Total remission was achieved in three treated lesions. The other lesions did not achieve total remission yet, but their sizes are diminishing. The differences observed in treatment outcome may be related with lesion features, adjuvant treatments and/or treatment schedule.

RESUMO

This study was aimed to work out a simple, applicable, sensitive and specific protocol for the identification of phosphoproteome. Isotope-labeling, two-dimensional electrophoresis, autoradiography and so on were used to establish a phosphoproteome map of mice neurons, and then chemiluminescence Western blotting was utilized to detect three phosphoproteins PI3Kr3, MEK1 and PKCalpha selectively. The results of comparison showed that the blots of PI3Kr3, MEK1 and PKCalpha on autoradiography map were almost identical with the blots of PI3Kr3, MEK1 and PKCalpha on chemiluminescence Western blotting maps. So this protocol based on isotope labeling and chemiluminescence Western blotting methods has proven to be sensitive and specific in the identification of phosphoproteome.

<p><b>BACKGROUND</b>Although 32P-glass microspheres (32P-GMS) have been used in internal radiotherapy for malignant tumors, it has been one of the key obstacles to improve the effect of radiotherapy. We investigated the cellular and hypersensitive effect of combined use of low dose of cisplatin and interstitial injection of 32P-GMS on mouse solid tumor S180.</p><p><b>METHODS</b>The mice with solid tumor S180 were randomly divided into four groups (controls, cisplatin therapy, 32P-GMS therapy and combination therapy). The specimens of the mice were sectioned two weeks after treatment and weighed. The death rate of tumor cells and the inhibition rate of tumor were calculated respectively. The cell cycle and apoptosis rate were evaluated with flow-cytometry. The ultrastructural changes of the four groups were observed by a transmission electron microscope. The data were analyzed by the chi-square test.</p><p><b>RESULTS</b>The growth of tumor was slower in the combination therapy group than in the simple therapy groups by macrography. The inhibition rate and the death rate of tumor cells of the combination therapy group were significantly higher than those of the control group and the other two simple therapy groups (P < 0.05). More cell damages were displayed in the combination therapy group than in the other groups under the light and electronic microscope.</p><p><b>CONCLUSION</b>Low-dose cisplatin combined with interstitial injection of 32P glass microspheres could be used as an effective hypersensitive regimen for the internal radiotherapy of mouse solid tumor S180.</p>

Interference of three dominant weed extracts viz., Ageratum conyzoides L., Melilotus indica All. and Parthenium hysterophorus L. were examined on seed germination, seedling growth, and nutrient uptake (32P and 65Zn) in three different varieties (PD-10, PD-12 and PB) of paddy (Oryza sativa L.). Among the three different varieties irrespective of weed extracts, PD-10 and PD-12 were resistant and PB was susceptible in terms of seed germination, radicle length and plumule dry weight; and PD-12 and PB were resistant and susceptible, respectively, in terms of plumule length and total seedling dry weight. A. conyzoides caused maximum reduction in seed germination and M. indica in seedling growth in different varieties of paddy. The weed extracts interfered in uptake of both 32P and 65Zn and there was a gradual decrease in uptake of both nutrients with increasing concentration of extracts in both root and shoot. The uptake of 32P and 65Zn was more inhibitory with the extracts of A. conyzoides and M. indica, respectively in different varieties. The inhibition in seed germination, seedling growth and nutrient uptake may be due to the presence of phenolics and other secondary metabolities. The phenolics such as gallic, vanillic, protocatechuic and p-hydroxybenzoic acids were identified from these weed extracts.

Se realizó un estudio retrospectivo de 279 pacientes con policitemia vera (PV) de los cuales 72 (25,8 por ciento) fueron tratados con 32P con un tiempo de seguimiento entre 4 y 28 años. La edad promedio fue de 61 años al recibir el primer tratamiento. El control hematológico se obtuvo en el 95,8 por ciento de los casos (n=69). La media de duración del control hematológico fue de 27,8 meses. La media de la dosis total utilizada por paciente fue de 9.2 ± 8,9 mCi. En 34 pacientes (54,1 por ciento) fue necesaria la administración de más de una dosis del radiofármaco durante el tiempo de evolución de la enfermedad. Del total de pacientes tratados, 5 (6,9 por ciento) desarrollaron una leucemia aguda entre los 9 y 25 años posteriores a la primera administración y 3 (4,1 por ciento) desarrollaron otras malignidades después de más de 15 años del diagnóstico. La mediana de supervivencia postratamiento fue de 10,1 años y la causa fundamental de muerte fueron los accidentes vasculares. No hay dudas de que el 32P constituye un método terapéutico conveniente y eficaz, fundamentalmente en pacientes mayores de 60 años, que logra una excelente calidad de vida y prolonga la supervivencia con un mínimo de reacciones adversas y un costo moderado

La braquiterapia o radioterapia intersticial cerebral inicia con Leksell en 1951. Ha evolucionado con los aportes de Backlund y su amplio estudio del efecto del p32 en craneofaringiomas quísticos, complementándose esta modalidad terapéutica con radio-gamma-cirugía cuando el componente sólido de la lesión así lo ameritaba. La experiencia es transmitida a través de algunos de los autores (JFDC y RG) a Venezuela donde se continuó y donde se siguen aportando modificaciones, con la utilización de otro radioisótopo, el I131 y ampliando su aplicación a otros tipos de tumores quísticos como los de la región pineal

Exposed to neutron flow, the phosphorus implanted TiNi alloy gets radioactive. This radioactive material is used in vascular stent for prevention and cure of restenosis. Phosphorus implantation is carried out in a plasma immerged ion implantation system, and the dose of phosphorus implantation is in the range of 2-10 x 10(17) cm-2. After ion implantation, the alloy is exposed to the slow neutron flow in a nuclear reactor, the dose of the slow neutron is 1.39-5.88 x 10(19) n/cm2. The radioactivity of the TiNi alloy was measured by liquid scintillation spectrometry and radio-chromic-film dosimetry. The result shows that whether the phosphorus is implanted or not, the TiNi alloy comes to be radioactive after exposure to neutron flow. Just after neutron irradiation, the radiation dose of phosphorus implanted TiNi alloy is about one hundred times higher than that of un-phosphorus implanted TiNi alloy. The radiation difference between phosphorus and un-phosphorus implanted alloy decreases as time elapses. Within three months after neutron irradiation, the average half-decay period of phosphorus implanted TiNi alloy is about 62 days. The radiation ray penetration of phosphorus implanted TiNi alloy is deeper than that of pure 32P; this is of benefit to making radiation uniformity between stent struts and reducing radiation grads beyond the edge of stent.

Protein tyrosine phosphorylation and dephosphorylation are important in the regulation of cell proliferation and signaling cascade. In order to examine whether phosphatase activity of CPTP1 and HPTP1B, typical nontransmembrane protein tyrosine phosphatase, could be controlled by phosphorylation, affinity-purified PTPs were phosphorylated by CKII and p56lck in vitro. Phosphoamino acid analysis revealed that CPTP1 was phosphorylated on both serine and threonine residues by CKII, and tyrosine residue by p56lck. Phosphatase activity of CPTP1 was gradually increased by three-fold concomitant with phosporylation by CKII. Phosphorylation of HPTP1B by CKII resulted in quick two-fold enhancement of its phosphatase activity within 5 min of incubation and remained in that state. In the presence of CKII inhibitor, heparin or poly(Glu.Tyr), both phosphorylation and enhancement of phosphatase activity of CPTP1 and HPTP1B were mostly blocked. p56lck catalyzed tyrosine phosphorylation of CPTP1 and HPTP1B was only observed by inhibiting the intrinsic tyrosine phosphatase activity. Taken together, these results indicate that CPTP1 or HPTP1B possesses a capability to regulate its phosphatase activity through phosphorylation processes and may participate in the cellular signal cascades.

With the purpose studying the effectivity of an intratumoral single dose of chromic [(32)P] phosphate with great particles for the treatment of solid tumors, studies of biolimination, biodistribution and therapeutic action were carried out. Only for comparative purpose, similar studies were undertaken using a solution of sodium [(32)P] orthophosphategelatine. The results show that when sodium [(32)P] orthophosphategelatine is used, the percentage of total elimination is (85.90+8,70) per cent with a higler percentage in urine (64.50+13.70) per cent than in faeces (21.40+4.50) per cent. In biodistribution studies, the greater percentage is found in bone (15.54+2.21) per cent while only a (2.51+0.39) per cent remains in the tumor. When great particles chromic [(32)P] phosphate was intratumorally injected, we determined that the total elimination is equal (36.28+6.27) per cent, finding a higler amount in faeces (29.44+5.26) per cent than in urine (6.84+2.21) per cent. Biodistribution studies demonstrated that (49.82+5.41) per cent remains in the tumor and (9.63+4.89) per cent of the injected activity is found in the liver. On the other hand, when therapeutic action was evoluted, we observed that the percentage of tumor regression (P.T.R) is 52.0 per cent for the tumors injected with chromic [(32)P] phosphate and 0.0 per cent for those injected with sodium [(32)P] orthophosphate-gelatine. These results show that the great particles colloid of chromic [(32)P] phosphate is not safe enough for the tratment of solid tumors, since it is mobilezed from the injection point, delivering a high dose to the whole organism.

In order to study the effect of nutrition on the onset of disturbances in Wistar rat pancreatic beta cells, we compared the effects of a low protein diet (8 per cent protein) and a normal protein diet (25 per cent protein) supplied to the dams (6 in each group) during the first 12 days of lactation. The parameter evaluated was the beta cells phosphate flush in response to stimulatory concentration of glucose (16.7 mM) of isolated islets of Langerhans from 60-day old pups. Using a collagenase digestion technique, islets were isolated from the pups and the 32P fractional outflow rate (FOR) of the beta cells was used as a metabolic index in both experimental groups (N = 36). We observed that although the weights of the pups of the two groups were not significantly different at 60 days of age (control = 186 +/- 18 g; undernourished during lactation = 179 +/- 19 g), the typical phosphate flush response (FOR = 2.4 +/- 0.4 per cent /min) to a stimulatory glucose concentration (16.7 mM) was abolished in the rats from undernourished mothers. Our data are consistent with the hypothesis that undernutrition may be an important cause of diabetes mellitus type II

RESUMO

A simple procedure was developed for spotting blood samples directly onto nylon membrane filter, without the necessity to treat samples with pronase or proteinase K, followed by hybridizing with 32P-labelled DNA probe, pUNK1-45. This probe detected specifically P. falciparum DNA and did not cross react with DNA from man, P. knowlesi, P. chabaudi or P. cynomolgi. The probe was sensitive to detect a parasitemia of 0.001% in 20 microliters of blood.

RESUMO

Se exponen los resultados obtenidos en el protocolo de tratamiento de la policitemia vera (PV) iniciado en 1967 por el grupo de estudio de la PV (GEPV). Se informa que después de más de 10 años de investigación, el GEPV se encontró frente a un dilema terapéutico. Se inicia que los casos tratados con flebóticas en los primeros 4 años; mientras que los pacientes tratados con clorambucil o p32 presentaban predominantemente otras enfermedades malignas, fundamentalmente leucemia aguda, después del cuarto año de tratamiento. Se señala que la búsqueda de una droga mielosupresora con débil potencialidad mutágena, se hizo evidente. Se ensaya, hace aproxidamente 5 años, con la hidroxiurea y hasta ahora todo parece indicar que la misma no produce complicaciones malignas superiores a las observadas con el tratamiento con flebotomías, aunque es necesaria una terapéutica a más largo plazo para poder hacer conclusiones definitivas. Se ha iniciado un nuevo protocolo hace 2 años, por un grupo de hematológos franceses, y aún no hay información sobre los resultados. Se expone de manera breve la experiencia de nuestra institución, en 17 años de trabajo con 56 pacientes con PV. Se expresa que el tratamiento con p32 y flebotomías, en pacientes mayores de 65 años, ha proporcionado una rápida y prolongada recuperación de estos enfermos; éste sería el indicado en los pacientes menores de 65 años sin factores de riesgo de trombosis, mientras que el tratamiento con HU a largo plazo, quedaría para los enfermos menores de 65 años pero que poseen estos factores de riesgo
