ABSTRACT

Objective To investigate the effects and the mechanism of tirofiban administration in myocardial ischemical reperfusion injury(MIRI) in patients with acute myocardial infarction(AMI) undergoing primary percutaneous coronary intervention(PCI). Method The study included 158 STEMI Patients who accepted primary PCI therapy and were randomly (random number) divided into two groups: tirofiban administration group and control group. Incidence of MIRI during PCI, Correct TIMI frame count(CTFC), ST segment resolution(STR), peak value and peak time of MB isoenzyme of creatine kinase( CK-MB), and incidence of major adverse cardiac events (MACE) during 30 days postoperation in both groups were measured. Results Tirofiban administration group was superior to control group in terms of incidence of MIRI, CTFC, STR, peak value and peak time of CK-MB, and incidence of MACE during 30 days postoperation (P < 0.05). Multiple factor logistic regression analysis indicated that intravenous tirofiban administration before primary PCI was the independently protective factor for MIRI. Conclusions Intravenous tirofiban administration in patients with STEMI before primary PCI can significantly decrease the incidence of MIRI,reduce myocardial damage and improve the prognosis.

ABSTRACT

OBJECTIVE:To study the protective effect of ethyl acetate extract from Japanese Artactylodes Rhizome on myocardial ischemical reperfusion injury in rats and its possible mechanism. METHODS:50 Wistar rats were divided randomly into 5 groups:sham group, model group, medicine (ethyl acetate extract from Japanese Artactylodes Rhizome at dosage of 200mg?kg-1 and 400mg?kg-1,respectively) group and verapamil(positive medicine) group. Rats in medicine group, model group and positive group were subjected to myocardial ischemia for 40min followed by reperfusion for 30 min, then muscular tissues were sampled for the determination of the activity of GSH-Px, the content of mitochondrial calcium (Mit-Ca2+),and the activities of Na+-K+-ATPase and Mg2+-ATPase in myocardium. RESULTS:As compared with model group, in the group treated with ethyl acetate extract from Japanese Artactylodes Rhizome 30min after reperfusion, the activity of GSH-Px was significantly higher,the content of myocardial mitochondrial calcium was significantly lower,and the activities of Na+-K+-ATPase, Mg2+-ATPase were significantly higher.CONCLUSION:The protective effect of ethyl acetate extract from Japanese Artactylodes Rhizome on myocardial ischemical reperfusion injury might be achieved through enhancing the activity of myocardial GSH-Px, lowering the content of myocardial mitochondrial calcium and enhancing the activities of Na+-K+-ATPase and Mg2+-ATPase of cellular membranes.

ABSTRACT

OBJECTIVE:To observe the effect of Bilobalide on ischemical reperfusion injury in orthotopic liver transplantation model rats and to discuss its protective effect on ischemical reperfusion injury of rats undergoing liver transplantation.METHODS:The model of orthotopic liver transplantation was established by in rats by setting up the bridge between portal vein and left renal vein and building blood bypass by inlaying of canal in the inferior vena,after modeling,the rats were treated with Bilobalide,then levels of NO(active medium of blood vessel)and ET1 before ischemia and at 10 min,30min and 2h after reperfusion were determined by nitratase reduction method;serum levels of ALT,AST,ALP,LDH enzymology,ATP and MDA in the liver tissues were determined as well.The hepatic tissue sample was taken at 2h and fixed with formalin to be made into specimen for observation of the ultrastructure of liver cells and hepatic lobules under electron microscope.RESULTS:After being treated by Bilobalide,serum NO level was elevated and the pathological elevated levels of ALT,AST and LDH were brought down,the microcirculation in ischemical reperfusion injury rats was improved and the extent of damage in the ultrastructure of liver cells was lessened in rats.CONCLUSION:Bypass of portal vein and inferior vena cava prior liver transplantation is helpful for the prevention of ischemical reperfusion injury of liver.The balance of NO/ET1 is possibly a factor influencing the blood flow in the microcirculation of transplanted liver.Bilobalide was proved to be of protective effect on ischemical reperfusion injury in rats.

ABSTRACT

OBJECTIVE:To study the protective effect of ethyl acetate extract from Oenanthe stolonifera against myocardial ischemia and ischemia reperfusion injury in rats.METHODS:Acute myocardial ischemia and ischemical reperfusion injury models were prepared by suture-occluded method.5 min before the onset of ischemia,the rats were injected with ethyl acetate extract from O.stolonifera,then the dynamic ECG change of rats was observed,and the arrhythmia caused by myocardial ischemia and the myocardial infraction size,the arrhythmia induced by myocardial ischemia-reperfusion,the activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in plasma were determined.RESULTS:Ethyl acetate extract from O.stolonifera significantly inhibited myocardial ischemia-induced arrhythmia and decreased the myocardial infarction size,significantly inhibited arrhythmia induced by myocardial ischemia-reperfusion,enhanced SOD activity and decreased the content of MDA in plasma.CONCLUSION:The protective effect of ethyl acetate extract from O.stolonifera against myocardial ischemia and ischemical reperfusion injury in rats is possibly associated to its anti-peroxidation effect.

ABSTRACT

Objective To investigate the protective mechanism of ischemic preconditioning for myocardial ischemic reperfusion injury (MI/RI) rats. Methods Eighteen male Sprague–Dawley (SD) rats were randomly separated into Sham operation,ischemic reperfusion (IR) and ischemic preconditioning (IP) groups (6 rats in each group). Peripheral blood and cardiac muscle were collected after the establishment of the above 3 animal models. ELISA was employed to determine proinflammatory cytokines TNF-? and IL-1? production in sera of these animals. RT-PCR and Western blot analysis was used to assay the transcriptional level and protein level of Toll-like receptor 4 (TLR4) in cardiac muscle tissue with pathological change,respectively. ABSTRACT

OBJECTIVE:Approach to the protective effect of traditional Chinese medicine that invigorating vital energy and promoting blood flow on model of renal ischemical reperfusion injury of rats.METHODS:The rats were divided into control group,model group and medication group(traditional Chinese medicine that invigorating vital energy and promoting blood flow),the medication group were given prevention administration for7days;model of renal ischemical reperfusion injury of rats were established.The contents of complement C3and nitric oxide(NO)in serum and renal tissue and the pathological changes of renal tissues in each group were compared.RESULTS:Levels of complement C3and NO in serum and renal tissue were increased in the model group and there were obvious pathological changes in renal tissues;compared with the model group,there was a decrease in the contents of complement C3and NO in serum and renal tissues and there was an obvious improvement in the renal tissues in the medication group.CONCLUSION:The traditional Chinese medicine that invigorating vital energy and promoting blood flow has a protective effect on the model rats with renal ischemical reperfusion injury.

ABSTRACT

OBJECTIVE:To study the protection of XNSK on renal ischemical reperfusion injury model rats.METHODS:A total of60rats were randomized into sham operation group(without occlusion of renal arteries of both kidneys and admin?istered with0.1%dimethyl sulfoxide normal saline),control group(0.1%dimethyl sulfoxide normal saline),positive control group(administered with Shenfukang capsules)and XNSK(high,medium and low dose subgroups)group.Levels of serum creatinine(Scr),urea nitrogen(BUN)and malonaldehyde(MDA)in renal tissue,hemocuprein(SOD),active oxygen(ROS)and nitrogen monoxidum(NO)were determined respectively meanwhile the pathomorphology changes of tissues were observed.RESULTS:As compared with the control group,levels of Scr,BUN,MDA,ROS and NO in all the3(high,medium and low doses)XNSK subgroups(which were administered with drugs for7days before operation and undergone reperfusion once after operation)had a reduction while SOD level had an increase.And there was a mitigated pathomorphology change in nephridial tissues(P

ABSTRACT

Aim To observe the protective effect and possible mechanisms of Total Flavone of Abelmoschus Manihot L medic(TFA) on myocardial reperfusion injury in rabbits,when it was given as a preconditioning inducer.Method Different doses of TFA were perfused intravenously lasting for 5 min,followed by a 10 min break before 30 min ischemia and 60 min reperfusion in rabbits.Using this preconditioning method,we observed the histo-pathological changes of myocardium,the content of MDA,the activities of SOD and GSH-PX in plasma,and also,the expression of ICAM-1 in myocardium.Results TFA(16,8 and 4 mg?kg~(-1)) significantly ameliorated myocardium ischemical reperfusion injury.The activities of SOD and GSH-PX in plasma were enhanced,while the MDA level was decreased.Meanwhile,the expression of ICAM-1 mRNA in myocardium was down-regulated.Conclusion Pharmacological preconditioning of TFA could protects myocardial ischemic reperfusion injury in rabbits by inhibiting lipid peroxidation and myocardium inflammatory reaction.
