ABSTRACT

AIM: To evaluate the bioequivalence of two kinds of amlodipine besylate tablets in Chinese healthy subjects under fasting and fed conditions. METHODS: Twenty-four healthy subjects were enrolled, and a random, open, single-dose, two preparations, two sequences and double-crossover design was used to give the test or reference preparations under fasting and postprandial conditions. The concentration of metformin in plasma was detected by LC-MS/MS, and the main pharmacokinetic parameters were calculated to evaluate the bioequivalence. RESULTS: In fasting state, the mean of C

ABSTRACT

OBJECTIVETo study the expression of telomerase reverse transcriptase (hTERT) and bcl-2 in ameloblastoma (AB), METHODS: hTERT mRNA in 54 cases of AB (primary AB 31 cases, recurrent AB 17 cases, malignant AB 4 cases) and 7 cases of oral normal mucosa was detected by in situ hybridization, and bcl-2 by S-P method.

RESULTSThe expression of hTERT mRNA was negative or weak in normal oral mucosa (14.3%), moderate or strong in AB (94.4%). There was a significant difference in these two groups (P < 0.001). The difference between the expressions of hTERT in primary, recurrent and malignant AB was significant (P < 0.05). The positive ratio of bcl-2 in AB and normal oral mucosa was respectively 88.0%, 44.4%. There was a significant statistical difference in these two groups (P < 0.001). hTERT mRNA was stronger in recurred or malignantly transformed AB (P < 0.05).

CONCLUSIONThe expression of hTERT and bcl-2 is stronger in recurred or malignantly transformed AB, and it could be used as an indicator of AB prognosis. Telomerase activity and bcl-2 expression play an important role in genesis and development of AB.

ABSTRACT

OBJECTIVEThis study was intended to determine the salivary protein factors related to adult periodonititis.

METHODSTwenty-five patients with adult periodontitis (AP group) and twenty-five normal controls (NC group, 25) were adopted in the study. Salivary protein composition in unstimulated whole saliva samples obtained before and after basic treatment was analyzed by SDS-PAGE.

RESULTSThe volume of proteins with 66 kD, 18 kD, 15 kD, 13 kD in AP group before treatment were all remarkably higher than those in NC group. In addition, 51 kD, 34 kD, 19 kD, 17 kD protein bands were more frequently observed in AP group by comparison with NC group. However, after initial therapy, all the variables mentioned above decreased remarkably.

CONCLUSIONThe results demonstrated that protein bands with 18 kD, 15 kD, 13 kD might be closely related to the occurrence and recovery of periodontitis, and serum-derived proteins in saliva increased in patients with adult periodontitis.

ABSTRACT

OBJECTIVETo study hTERT mRNA expression in ameloblastoma (AB) and odontogenic keratocyst (OKC) and to investigate genesis, development and biological characteristics of AB and OKC.

METHODShTERT mRNA expression in 54 cases of AB, 16 cases of OKC, 7 cases of oral normal mucosa was detected by in situ hybridization.

RESULTSThe positive rates of hTERT mRNA in AB, OKC, and oral normal mucosa were 94.4% (51/54), 87.5% (14/16), 1/7, respectively. There was significant statistical difference (P < 0.001). About clinicalpathology of AB, there was no difference for hTERT expression (P > 0.05) hTERT mRNA was positive in peripheral and stellate reticula cells.

CONCLUSIONS(1) hTERT activity plays an important role in genesis, development of AB. (2) hTERT positive rate is related to cell differentiation and clinical biological behavior.

ABSTRACT

OBJECTIVEThe aim of this study was to investigate the features of mast cell subtypes and relationship between mast cells and pathogenesis of oral lichen planus.

METHODSMast cells in the tissue of oral lichen planus were stained with Alcian blue/safranine in order to demonstrate mature and immature mast cells, and the constituent ratio of these two subtypes was calculated. Afterwards, LUZEX-F image pattern analysis system was applied to demonstrate the size of these two subtypes and their morphological features.

RESULTSThe constituent ratio of mature and immature types of mast cells in the tissue of oral lichen planus was significantly different from that in the normal tissue (P < 0.01), and such difference was also demonstrated among different types of this malady, including papule-, net- and erosion-type of oral lichen planus (P < 0.05). There was significant difference between the sizes of mature and immature mast cells (P < 0.01) and also the sizes, amount and distribution of the plasma granules within these two mast cells.

CONCLUSIONIt could be implied that mast cells might play a role in the pathogenesis of oral lichen planus.

ABSTRACT

Objective:To investigate the effects of Candida albicans(C. albicans) on the cell cycle distribution of oral epithelial KB cells in vitro. Methods:KB cells at 3?106/ml were seeded and cultured for 48 h, then the bacterial filament and the spore of C. albicans at 102 CFU/ml were added to the cultures respectively. KB cell culture without C. albicans was used as the control. After 48 h culture cell suspensions were prepared and subjected to FACS flow cytometry for detecting the cell cycle distribution. Results:Filament of C. albicans decreased the percentage of G0/G1 cells, increased G2/M cell population and proliferation index of the cells(P

ABSTRACT

Objective:To investigate the expressions of epidermal growth factor receptor(EGFR) and C-erbB-2 in two salivary adenoid cystic carcinoma cell lines with different metastatic potential. Methods:Salivary adenoid cystic carcinoma SACC-83 cell line and its lung metastatic cell line SACC-LM were cultured in vitro, their cytoplasm and membrane samples were extracted, and the expressions of EGFR and C-erbB-2 were detected by Western blot, the results were quantitatively analyzed by FluorChem V2.0 software. Results:EGFR expression level in cell membrane was higher than that in cytoplasm(P0.05) in both cell lines. Conclusion:Higher EGFR expression in cytoplasm and higher C-erbB-2 expression may be related to the metastatic potential of SACC-LM cells.

ABSTRACT

Objective: Our aim was to analyze the relationship between the concentration of some minerals in saliva and periodontitis.Methods:Twenty-five patients with adult periodontitis (AP group), 25 age and sex matched healthy people(N group) were selected. Unstimulated whole saliva samples were collected and qualified for the concentration of TCa,P,Mg2+, Na+,K+, and Cl-. Clinical parameters on periodontitis were also assessed for the patients. All the aforementioned indices were measured repeatedly at the end of the fourth week after initial therapy. Results:The clinical condition of the patients improved significantly after treatment. The level of saliva Ca elevated before and after treatment in AP group than that in N group. Before treatment, the concentration of Na+ also increased significantly, but it decreased to normal level after initial therapy. The findings also indicated the positive relationship between the concentration of salivary Na+ and pocket probing depth as well as gingival index before treatment. Conclusion:The elevated level of saliva Ca is a sign of periodontitis and may help the occurrence and progress of periodontitis. The level of saliva Na+ may indicate the pocket depth and degree of gingivitis. The saliva Na+ is closely associated with the clinical condition of periodontitis.
