ABSTRACT

Objective:To explore the correlation between Traditional Chinese Medicine (TCM) syndrome types of Sj?gren syndrome (SS) and blood test parameters, immunological function and disease activity. Methods:A retrospective cross-sectional study was conducted. The clinical data of 242 SS inpatients in the Rheumatology and Immunology Department of Jiangsu Province Hospital of TCM from February 2021 to June 2022 were analyzed retrospectively. We compared the general data (gender, age, course of disease, BMI), blood parameters [WBC, hemoglobin (Hb), PLT, neutrophil count (NEUT), lymphocyte count(LYMPH), neutrophil/lymphocyte ratio (NLR)], immunological indicators (globulin, IgG, IgA, IgM, rheumatoid factor (RF), anti-SSA antibody, anti-SSB antibody, anti-Ro-52 antibody) .The distribution difference of disease activity [Disease Activity Index of Sjogren's syndrome (ESSDAI) and Patient Report Index of Sjogren's syndrome (ESSPRI)], the correlation between each syndrome type and blood routine parameters, immunological indicators and inflammatory indicators was analyzed by binary logistic regression. Results:They were divided into 82 cases of qi yin deficiency syndrome, 61 cases of yin deficiency and fluid deficiency syndrome, 59 cases of yin deficiency and blood stasis syndrome, 32 cases of yin deficiency and heat toxin syndrome, and 8 cases of other syndrome types. Because the number of other syndrome types was small, they were not included in this study. Logistic regression analysis showed that the positive rate of anti SSA antibody was negatively correlated with IgM [ OR (95% CI)=0.570 (0.407, 0.798)] ( P<0.01). The positive rates of anti SSB antibody and anti Ro-52 antibody were negatively correlated with LYMPH [ OR (95% CI)=0.445 (0.223, 0.886), 0.457 (0.224, 0.932), respectively] ( P<0.05). The positive rates of anti SSB antibody and anti Ro-52 antibody were positively correlated with IgG [ OR (95% CI)=1.171 (1.034, 1.325), 1.159 (1.014, 1.325), respectively] ( P<0.05). Qi Yin deficiency syndrome was positively correlated with WBC [ OR (95% CI)=2.590 (1.120, 5.987)] ( P<0.05), and negatively correlated with LYMPH [ OR (95% CI)=0.090 (0.017, 0.470)] and IgA [ OR (95% CI)=0.728 (0.553, 0.959)] ( P<0.05). Yin deficiency and fluid deficiency syndrome were negatively correlated with PLT [ OR (95% CI)=0.991 (0.984, 0.998)], ESSPRI [ OR (95% CI)=0.705 (0.506, 0.983)], ESSDAI [ OR (95% CI)=0.716 (0.534, 0.960)] ( P<0.05). Yin deficiency and blood stasis syndrome was positively correlated with IgA [ OR (95% CI)=1.184 (1.028, 1.363)] ( P<0.05), and negatively correlated with anti SSB antibody positive rate [ OR (95% CI)=0.247 (0.093, 0.659)] ( P<0.05). Yin deficiency heat toxin syndrome was positively correlated with IgA [ OR (95% CI)=1.368 (1.037, 1.803)] ( P<0.05), and negatively correlated with anti SSB antibody positive rate [ OR (95% CI)=0.278 (0.085, 0.909)] ( P<0.05). Conclusion:The level of immunoglobulin, inflammatory index and disease activity of yin deficiency and blood stasis syndrome and yin deficiency and heat toxin syndrome are high, and blood system damage and exocrine gland disease are easy to occur, which can provide clinical basis for the combination of disease and syndrome differentiation and treatment of SS.

ABSTRACT

Objective To investigate the immunological functions of heat shock protein 40 kDa of Schistosoma japonicum (SjHSP40). Methods The homology of the SjHSP40 protein sequence was analyzed and the B and T cell epitopes of SjHSP40 were predicted using bioinformatics tools. The full-length SjHSP40 gene was amplified using a PCR assay, and cloned into the prokaryotic expression vector pGEX-6P-1, which was transformed into Escherichia coli BL-21. The protein expression was induced with isopropyl β-D-thiogalactoside (IPDG), and then, the recombinant protein was purified with glutathione-sepharose 4B resin to yield the fusion protein GST-SjHSP40, which was checked with SDS-PAGE and Western blotting. Following immunization with GST-SjHSP40, the serum levels of anti-SjHSP40 IgG antibody and IgG1 and IgG2a subtypes were detected in BALB/c mice using ELISA. In addition, the effect of SjHSP40 on CD4+ T-cell subset differentiation was examined using flow cytometry. Results SjHSP40 contained 7 potential B cell epitopes and multiple T cell epitopes (CTL epitopes and Th epitopes). The prokaryotic expression plasmid pGEX-6p-1-SjSHP40 was successfully constructed, and the fusion protein GST-SjHSP40 was obtained following IPDG induction and protein purification. Significantly higher serum levels of anti-SjHSP40 IgG, IgG1 and IgG2a antibodies were detected in mice immunized with GST-SjHSP40 than in other groups; however, SjHSP40 showed no remarkable effects on CD4+ T-cell subset differentiation. Conclusions SjHSP40 may induce specific humoral immune responses in mice; however, it does not affect the balance of Th immune responses. It is suggested that SjHSP40 may be a potential vaccine candidate.

ABSTRACT

The oncogenic mechanism of human papilloma virus (HPV) has been elucidated, and high-risk HPV (HR-HPV) infection may cause malignancies such as cervical cancer.HR-HPV infection of the cervix is very common, while only a few cases of such infection develop into malignancy. Therefore, the evaluation of oncogenic risk of cervical HPV infection is of great clinical significance.Persistent HR-HPV infection of the cervix is mainly caused by immunological dysfunction-induced failure of HPV elimination and supposed to be the precondition of cervical cancer, while the type of HPV is an important factor for carcinogenesis of epithelial cells.

ABSTRACT

Objective To explore the effects of the purified Plasmodium falciparum(P. f)antigen on the T lymphocytes of the peripheral blood of healthy people in vitro. Methods The peripheral blood mononuclear cells(PBMCs)isolated from healthy donators were cultured with purified P. f antigen(5μg/ml)and normal RBC(nRBC)antigen(5μg/ml),in addition, the group in which only IL-2 added was set as a negative control. After cultured for 12 d,the corresponding antigens were used to re-stimulated the activated T cells in the two stimulated groups for 20 h. Then the cells were collected, the proliferations of the T cells labeled by CFSE were analyzed, and the secretion condition of IL-4 and IFN-γof the cells were detected by flow cytome-try. Results After amplification, the proliferative index(PI )of CD4+T cells that were stimulated with P. f antigen was signifi-cantly higher than those in the nRBC antigen group and the negative control group(both P0.05). Meanwhile, the percentages of the CD4+T cells se-creting IL-4+in the P. f antigen group was significantly higher than those in the other two groups(both P0.05). Conclusion The purified P. f anti-gen could effectively stimulate the proliferation of CD4+T cells in peripheral blood of healthy people, and the latter can play a role in immunoregulation through secreting IL-4 preferentially.

ABSTRACT

Objective To investigate the effect of Kanglaite injection (KLT) on immunological function of rat models with Lewis lung carcinoma. Methods Forty C57BL/6 mice were used to establish Lewis lung carcinoma models and divided randomly into the high dose(25 mL/kg), middle dose (12.5 mL/kg) and low dose (6.25 mL/kg) of KLT groups and model group(n=10). The mice in the KLT groups were sacrificed after injecting corresponding dose of KLT with intraperitoneal injection for 14 d. No treatment was performed on the rats in model group. The body weight, tumor and spleen weight was weighed, then the ratio of tumor restriction and the index of spleen was calculated. MTT colorimetric method and ELISA were used to detected activity of T cell proliferation and expression of IL-2 in spleen. The expression of NF-κB and IκBα protein was detected by Western blot. Results The ratio of tumor restriction in the high, middle, low dose of KLT groups decreased gradually. The indexes of spleen of the high and middle dose of KLT groups were higher than those in the model group (P<0.05 or P<0.01).Compared with the model group, the activity of T cell proliferation in the high, middle, low dose of KLT groups and the expression of IL-2 in the high and middle dose of KLT groups was increased significantly (P<0.05 or P<0.01). The expression of NF-κB protein in the nuclei of high, middle, low dose of KLT groups increased dose-dependently, and the expression of NF-κB and IκBα protein in the cytoplasm decreased dose-dependently. ConclusionKLT could enhance immunological function by effecting T cell proliferation, expression of IL-2, NF-κB and IκBα, while restricting tumor growth in Lewis lung carcinoma models.

ABSTRACT

Objective: To investigate the effects of new Coriolus versicolor polysaccharide peptide (PSP) in enhancing the action and reducing the toxicity induced by cyclophosphamide (CTX) in mice bearing S180 sarcoma, analyze the related immunological mechanisms, and compare the effect of PSP2 (new polysaccharide peptide) with that of PSP1 (old polysaccharide peptide). Methods: S180 sarcoma xenografted mice models were established. The model mice were divided randomly into 8 groups, including model control group, CTX group, CTX plus PSP2 (200, 400, and 800 mg/kg) groups, CTX plus PSP1 400 mg/kg group, PSP2 400 mg/kg group, PSP1 400 mg/kg group. The normal blood cells and the nucleated marrow cells were counted. The survival time, the inhibitory rate of tumor growth, the index of thymus (thymus gland weight/body weight), spleen index (spleen weight/body weight) were measured. The subtypes of T lymphocytes in spleen were detected by flow cytometry. Results: PSP2 800 mg/kg combined with CTX prolonged the life span of the S180 sarcoma xenografted mice (P < 0.05). PSP2 and PSP1 alone obviously inhibited the growth of sarcoma, respectively (P < 0.05). PSP2 and PSP1 remarkably enhanced the ability of CTX to inhibit the growth of sarcoma as well (P < 0.05 or 0.01). PSP2 and PSP1 attenuated the immunological inhibition and marrow inhibition caused by CTX. PSP2 and PSP1 alone elevated the ratio of CD3+ CD4+/CD3+ CD8+ in mice bearing S180 sarcoma. The ratio tended to increase after combined treatment (PSP2 + CTX or PSP1 + CTX). Especially it increased remarkably after PSP2 400 mg/ kg combined with CTX (P < 0.05). Conclusion: PSP2 and PSP1 inhibited tumor growth, increased the action and reduced the toxicity of CTX. The mechanisms were related with enhancing the ratio of CD3+ CD4+/CD3+ CD8+ of spleen cells. PSP2 is effective than PSP1 in inhibiting tumor growth, enhancing the acion of CTX, and regulating the immunological function of nude mice.

Objective To investigate the anti-tumorigenesis function of DCN gene and its effect on the immunological funotions of mice bearing S180.Methods Twenty four mice bearing S180 were divided into three groups at random(eight mice every group),and then the normal saline group(negative control),pcDNA3.1(+)vector group and pcDNA3.1(+)-DCN group were set up for the experiment. They all started to inject next day by armpit. The volume of tumors were measured at the 5th day,7th day,9th day,11th day.The transformation rate of T lymphocytes were detected by means of MTT,the mRNA level of TGF-β by RT-PCR and the level of VEGF in serum by ELISA.Results The average volume of tumor in pcDNA3.1 (+)-DCN group were deflated to(2.219±0.105)cm3 comparing with the others(P<0.001);the transformation rate of T lymphocytes were increased significantly to(67.21±6.32)%(P<0.01,P<0.05);meanwhile it can decrease the mRNA level of TGF-β in serum, and depress the expression of VEGF markedly, whose absorbance were2.6175±0.099(P<0,01,P<0.05).Conclusion DCN gene could significantly inhibit the growth of tumor, and enhance the immunological function of mice bearing S180 in vivo.

ABSTRACT

[Objective] Using gene chip to detect the influence on gene expression due to replicate Heart-Qi Deficiency Syndrome on Wistar Rat caused by complex factor including slimming, forced swimming and large dosage of propanolol. [Methods] Every animal in experiment group took food according to 5g/100g body weight, and was forced to swim by taking weight according to its 5% body weight until sank beyond 10 seconds. From 17th day ig propanolol 24mg/Kg and lasted for 4 days. At 21st day we took out hearts and picked up general DNA to prepare probe, then cross with gene chip. [Results] Compared with control group, expression of gene NM-017329,NM-016997 and NM-030850 showed down regulation obviously, expression of gene NM-031345 and NM-024349 showed up-regulation significantly. [Conclusion] Repression of immunological function and damage of vascular endothelium may be the main mechanism resulting in Heart-Qi Deficiency Syndrome.
