ABSTRACT

BACKGROUND: Lawsonia intracellularis remains a problem for the swine industry worldwide. Previously, we designed and obtained a vaccine candidate against this pathogen based on the chimeric proteins: OMP1c, OMP2c, and INVASc. These proteins formed inclusion bodies when expressed in E. coli, which induced humoral and cellular immune responses in vaccinated pigs. Also, protection was demonstrated after the challenge. In this study, we established a production process to increase the yields of the three antigens as a vaccine candidate. RESULTS: Batch and fed-batch fermentations were evaluated in different culture conditions using a 2 L bioreactor. A fed-batch culture with a modified Terrific broth medium containing glucose instead of glycerol, and induced with 0.75 mM IPTG at 8 h of culture (11 g/L of biomass) raised the volumetric yield to 627.1 mg/L. Under these culture conditions, plasmid-bearing cells increased by 10% at the induction time. High efficiency in cell disruption was obtained at passage six using a high-pressure homogenizer and a bead mill. The total antigen recovery was 64% (400 mg/L), with a purity degree of 70%. The antigens retained their immunogenicity in pigs, inducing high antibody titers. CONCLUSIONS: Considering that the antigen production process allowed an increment of more than 70-fold, this methodology constitutes a crucial step in the production of this vaccine candidate against L. intracellularis.

ABSTRACT

Several pathogens and antibodies derived from serum or produced in tissues associated with the oral cavity are present in the oral fluid (OF). Considering the applicability of this alternative sample, recent studies in veterinary medicine have tested OF as a replacement for serum in diagnostic assays. The aim of this study was to standardize the immunoperoxidase monolayer assay (IPMA) to detect anti-Lawsonia intracellularis immunoglobulin A (IgA) and immunoglobulin G (IgG) in OF samples from experimentally infected pigs. Sixty-two pigs were divided into two groups: control (T1, n=30) and inoculated with L. intracellularis (T2, n=32). Blood, OF and fecal samples were collected at 0, 7, 14, 21, 28 and 42 days post-inoculation (dpi). Some adaptations of the standard technique for serum were made to IPMA for the detection of IgA and IgG in OF. The IPMA showed high specificity and sensitivity for serum samples and high specificity and moderate sensitivity for the detection of IgA and IgG in OF. There was high agreement between the results of serum IgG and OF IgA and IgG. Based on our results, oral fluid samples may be used for the evaluation and determination of anti-L. intracellularis antibodies in pigs, but not for individual diagnosis of swine proliferative enteropathy.(AU)


Vários patógenos e anticorpos derivados do soro ou produzidos em tecidos associados a cavidade oral estão presentes no fluido oral (FO). Considerando a aplicabilidade dessa amostra alternativa, estudos recentes em medicina veterinária têm testado o FO como substituto do soro para testes diagnósticos. O objetivo desse estudo foi padronizar a imunoperoxidase em monocamada de célula (IPMC) para a detecção de imunoglobulina A e imunoglobulina G anti-Lawsonia intracellularis em amostras de FO de suínos experimentalmente infectados. Um total de 62 suínos foram divididos em dois grupos: controle (T1, n=30) e inoculados com L. intracellularis (T2, n=32). Sangue, FO e amostras de fezes foram coletados aos 0, 7,14, 21, 28 e 42 dias após a inoculação (dpi). Algumas adaptações da técnica foram realizadas na técnica padrão da IPMC para a detecção de IgA e IgG. A IPMC demostrou alta especificidade e sensibilidade para amostras de soro e alta especificidade de moderada sensibilidade para a detecção de IgA e IgG em FO. Houve alta concordância entre resultados de detecção de IgG em soro com a IgA e IgG em amostras de FO. Baseado em nossos resultados, amostras de fluido oral podem ser usadas em avaliações e detecção de anticorpos anti-L. intracellularis em suínos, porém não de forma individual.(AU)

ABSTRACT

Porcine proliferative enteropathy (PPE) is one of the most common enteric diseases in growing and finishing pigs. PPE is characterized by reduced growth performance, accompanied or not by diarrhea. PPE is highly prevalent in several countries of the Americas, Europe and Asia, causing high economic losses in swine herds. The most common form of PPE control in pigs is antibiotic therapy. The objective of this study was to evaluate a new product based on tylosin injectable (Eurofarma Laboratórios S.A.) to control PPE in experimentally inoculated animals. Sixty 5-week-old pigs with mean weight of 9.5kg were divided into two experimental groups of 30 animals: medication and control. All pigs were challenged with Lawsonia intracellularis, the etiologic agent of PPE, on day zero. Fecal score, body condition score, and behavior were daily evaluated. Pigs were weighted on days -2, 13 and 21 of the experiment. Pigs in the Medication Group received tylosin injectable 13 days after inoculation, in three doses with a 12-hour interval between them. Pigs in the Control Group received injectable saline solution following the same protocol. In the Control Group, 23pigs presented with diarrhea before day 13. After day 13, the number of diarrheic animals in this group was reduced to 17. In the Medication Group, 26 pigs presented with diarrhea in the initial period, and in the period after medication, only 11 animals had diarrhea. The score of gross intestinal PPE lesions in the Medication Group was lower than that in the Control Group (p=0.031). The Medication Group also showed lower score for Lawsonia intracellularis antigen-labeling by immunohistochemistry compared with that of the Control Group (p=0.032), showing lower level of infection. These results demonstrate that tylosin injectable (Eurofarma Laboratórios S.A.), administrated in three doses (1mL/20kg) every 12 hours, was effective for the control of PPE in experimentally inoculated pigs.(AU)


Enteropatia proliferativa suína (EPS), causada pela bactéria Lawsonia intracellularis, é uma das doenças entéricas mais comuns em suínos de recria e terminação. A EPS caracteriza-se por redução no desempenho dos animais, acompanhada ou não por diarreia. É uma doença altamente prevalente em diversos países da América, Europa e Ásia, provocando elevados prejuízos econômicos nos rebanhos suínos. A forma de controle da EPS mais adotada em rebanhos suínos é a antibioticoterapia. O objetivo deste estudo foi avaliar um novo produto à base de tilosina (Eurofarma Laboratórios S.A.) na forma injetável para controlar a EPS em animais experimentalmente inoculados. Foram utilizados 60 leitões, de cinco semanas de idade, com peso médio de 9,5kg, divididos em dois grupos experimentais (n=30), medicados e não medicados. Todos os leitões foram desafiados com Lawsonia intracellularis no dia zero. Avaliações clínicas de escore fecal, escore corporal e comportamento foram realizadas diariamente além da pesagem individual dos animais realizada nos dias -2, 13 e 21 do experimento. Os leitões do grupo medicado receberam tilosina injetável 13 dias após a inoculação em três doses com intervalo de 12 horas cada. Já os leitões do grupo não medicado receberam solução salina injetável com o mesmo protocolo. O grupo não medicado apresentou 23 animais com diarreia antes do dia 13 e 17 após este período. No grupo medicado, 26 animais apresentaram diarreia previamente à medicação e apenas 11 após a medicação a partir do dia 13. Os leitões medicados apresentaram extensão de lesão macroscópica, caracterizada por espessamento de mucosa intestinal, menor em comparação com o grupo não medicado (p=0,031). A imunomarcação para Lawsonia intracellularis foi menor no grupo medicado (p<0,032), mostrando redução no grau de infecção por L. intracellularis nos animais medicados. Estes resultados demonstram que a tilosina injetável (Eurofarma Laboratórios S.A.) (1mL/20kg) em três doses, a cada 12 horas, foi eficaz no tratamento da enteropatia proliferativa suína em animais experimentalmente inoculados.(AU)

ABSTRACT

Porcine proliferative enteropathy (PPE) caused by Lawsonia intracellularis (LI) is a global cause for substantial economic losses in the swine industry. Here, we constructed live attenuated Salmonella typhimurium (ST) mutant strains expressing and secreting 4 selected immunogenic LI antigens, namely, optA, optB, Lawsonia flagellin (LfliC), and Lawsonia hemolysin (Lhly); the resultant recombinant strains were designated Sal-optA, Sal-optB, Sal-LfliC, or Sal-Lhly, respectively. Using the BALB/c mouse model, we demonstrate that mice vaccinated once orally, either with a mixture of all 4 recombinant strains or with an individual recombinant strain, show significant (p < 0.05) production of LI-specific systemic immunoglobulin (Ig) G and mucosal IgA responses compared to the Salmonella alone group. Upon restimulation of vaccinated splenocytes with the LI-specific antigens, significant (p < 0.05) and comparable production of interferon-γ responses are found in all vaccinated groups, except the Sal-Lhly group, which shows non-significant levels. Challenge studies were performed in C57BL/6 vaccinated mice. On challenge with the LI (10(6.9) 50% tissue culture infectious dose) 14 days post-vaccination, 20% (1/5) of mice in all vaccinated groups, except Sal-Lhly group, show the presence of the LI-specific genomic DNA (gDNA) in stool samples. In contrast, 40% (2/5) and 60% (3/5) of mice vaccinated with the Sal-Lhly strain and the attenuated Salmonella alone, respectively, were found positive for the LI-specific gDNA. Furthermore, 0% mortality was observed in mice vaccinated against the ST challenge compared to the 30% mortality observed in the unvaccinated control group. In conclusion, we demonstrate that the Salmonella-based LI-vaccines induce LI-specific humoral and cell-mediated immunities, and encompass the potential to offer dual protection against PPE and salmonellosis.

Porções de íleo terminal foram coletados de 100 suínos com sinais de doença gastrointestinal na área metropolitana de Bucaramanga, a fim de se estudar a eficiência do diagnóstico de enteropatia proliferativa suína (PPE) pela técnica de PCR aninha (PCRa) empregando sequências específicas (primers) para L. intracellularis: 16S ARN região (270pb) e sua correlação com achados clínicos e patológicos. Todas as amostras foram processadas para se determinar a associação entre positividade por PCR, os sinais clínicos, os achados de necropsia e as lesões histológicas. Cinquenta e seis por cento das amostras foram positivas para L. intracellularis pela PCRa. Só 2% exibiram resultados positivos pela técnica Warthin-Starry. Trinta e um de 100 animais com sinais de anorexia resultaram positivos para PCRa (P>0,05). Não houve associação (P<0,05) entre diarreia e queda no crescimento, bem como associação (P<0,05) entre achados anatomopatológicos e histológicos com PCRa positivas.(AU)


Fragments of terminal ileum were collected from 100 pigs at slaughter from Bucaramanga Metropolitan Area (Santander, Colombia), to study the efficacy of the diagnosis of porcine proliferative enterophaty (PPE) through the technique of nested polymerase chain reaction (PCRa), employing specific sequences (primers) for L. intracellularis: 16S ARN region (270pb) and his correlation with clinic and pathological findings. All samples were processed by standard histological methods and stained with a Warthin-Starry technique. All samples were processed to determinate the association between positive PCRa results, clinical signs and necropsy findings. 56% of the 100 samples were positive for L. intracellularis through PCRa technic. Only 2% exhibited positive results through Warthin-Starry technique. A total of 31 (100) animals with anorexic symptoms were associated with positive results from PCRa (P>,05). No associations (P<0.05) were observed between diarrhea and delayed growth. No associations (P<0.05) were observed between anatomopathological and histological findings with positive PCRa.(AU)

ABSTRACT

Lawsonia intracellularis infection on a horse farm in the Midwest region of Brazil is described. Thirty-nine foals a few days to months old from a herd with 300 horses, experienced diarrhea with variable characteristics and intensities, weight loss, hyperemic mucous membranes and dehydration. In foals 3 to 6 months of age, hypoproteinemia associated with submandibular edema were also common. Intestinal fragments of a 7-month-old foal were sent to an animal disease laboratory for diagnosis. The observed macroscopic lesions were hyperemic serosa, thickening of the intestinal wall with a corrugation, thickening of the mucosa folds and reduction of intestinal lumen. Histological analysis of the small and large intestine revealed enterocyte hyperplasia of the crypts associated with diffuse marked decrease in the number of goblet cells and positive L. intracellularis antigen labeling by immunohistochemistry. Three out of 11 animals of the same property were seropositive for L. intracellularis, demonstrating the circulation of the agent throughout the farm, but none were PCR positive in fecal samples. Based on clinical signs and pathological findings, the diagnosis of equine proliferative enteropathy was confirmed.


Descreve-se a infecção por Lawsonia intracellularisem uma propriedade na região Oeste do Brasil. Em um rebanho de 300 equinos, 39 potros com poucos dias de vida à 21 meses apresentaram diarreia de características e intensidades variáveis, com perda de peso e desidratação. Em potros com três a seis meses de idade, hipoproteinemia associada a edema submandibular também foram frequentes. Fragmentos intestinais de um potro de 7 meses foram enviados ao laboratório de patologia animal para diagnóstico. Na macroscopia foi observada hiperemia de serosa e moderado espessamento de parede intestinal. Na histologia do intestino delgado existia hiperplasia de enterócitos de criptas difusa intensa com redução marcante de células caliciformes e marcação positiva na imuno-histoquímica para L. intracellularis. Na sorologia de 11 animais da mesma propriedade, três foram positivos. Já a PCR foi negativa para todos os animais. Com base nos sinais clínicos e nos achados patológicos confirmou-se o diagnóstico de enteropatia proliferativa equina, associada a sorologia positiva que demonstrava circulação do agente na propriedade.

ABSTRACT

Proliferative enteropathy caused by Lawsonia intracellularis is one of the most common enteric diseases in pigs. The objective of this study was to determine the prevalence of serum antibodies against L. intracellularis in the general swine population of Korea from 2005 to 2008. In total, 8,008 swine serum samples obtained from 1,001 herds were tested. The samples were analyzed with an immunoperoxidase monolayer assay to detect anti-L. intracellularis antibodies. The overall 4-year average true prevalence was 40.0% (CI: 39.4 - 40.6%) at the individual animal level and 71.9% (CI: 70.3-73.4%) at the herd level.

ABSTRACT

The susceptibility of sparrows (Passer domesticus) and strains of mice (Swiss, BALB/c, C-57 and DB-A) to Lawsonia intracellularis infection was studied. Thirty-two sparrows were inoculated with pure culture of L. intracellularis and eleven received sham inoculum. Feces were collected on -1, 7, 14 and 21 days post infection (dpi) for detection of L. intracellularis by PCR. After 21 days, all sparrows were euthanized and the tissues processed for histology and immunohistochemistry (IHC). One hundred sixty mice of four different strains (n=40, per strain) were used. For each mouse strain, 16 animals received mucosa homogenate from a pig infected with L. intracellularis, 16 received pure culture of L. intracellularis and eight animals received sham inoculum. Two control and four inoculated mice from each group were euthanized on 7, 14, 21 and 28 dpi. Sections of intestine were collected for histologic analysis and IHC and pooled feces were collected for L. intracellularis PCR. None of the sparrows had any histologic lesions characteristic of proliferative enteropathy or antigen labeling by IHC. All sparrow fecal samples were negative by PCR. All mice strains studied had histopathological lesions typical of PE and IHC labeling consistent with L. intracellularis infection, especially those animals inoculated with pure culture. The most severe lesions were observed in DB-A and Swiss mice. Fecal shedding was detected in all mice strains, with peak at 14 dpi. We conclude that sparrows do not seem to be relevant in the epidemiology of L. intracellularis. The results showed variations in the lesions among the four mice strains used.


A susceptibilidade de pardais (Passer domesticus) e linhagens de camundongos (Swiss, BALB / C, C-57 e DB-A) à infecção por L. intracellularis foi testada. Trinta e dois pardais foram inoculados com cultura pura de L. intracellularis e onze receberam placebo. As fezes foram coletadas nos dias -1, 7, 14 e 21 após a infecção (dpi) para a detecção de Lawsonia intracellularis por PCR. Após 21 dias, todos os pardais foram eutanasiados e os tecidos processados para a realização da histologia e imuno-histoquímica (IHQ). Cento e sessenta camundongos de quatro linhagens diferentes (n=40, por linhagem) foram utilizados. Para cada linhagem de camundongo, 16 receberam homogeneizado de mucosa preparado a partir de um suíno infectado com L. intracellularis, 16 receberam cultura pura de L. intracellularis e oito animais receberam placebo. Dois camundongos controle e quatro camundongos inoculados de cada grupo foram sacrificados aos 7, 14, 21 e 28 dpi. Seções de intestino foram coletadas para análise histológica e IHQ e amostras de fezes foram coletadas para a realização da PCR para detecção de L. Intracellularis. Nenhum dos pardais apresentou lesões histológicas características da enteropatia proliferativa ou marcação positiva por meio da IHQ. As amostras de fezes dos pardais foram negativas na PCR. Todas as linhagens de camundongos estudadas tinham lesões histopatológicas típicas de enterite proliferativa e IHQ positiva para a infecção por L. intracellularis, especialmente aqueles animais inoculados com a cultura pura. As lesões mais graves foram observadas em camundongos DB-A e Swiss. A eliminação fecal foi detectada em todas as linhagens de camundongos, com pico 14 dpi. Conclui-se que os pardais não são relevantes na disseminação da L. intracellularis. Os resultados mostraram variações nas lesões entre as quatro linhagens de camundongos utilizadas, indicando o potencial risco que os camundongos representam na transmissão de L. Intracellularis.

PURPOSE: Porcine proliferative enteropathy (PPE) is known as one of the most important risk factors causing economic losses in swine industry worldwide. This study was conducted to evaluate the efficacy of a commercial oral attenuated Lawsonia intracellularis vaccine (Enterisol Ileitis) against PPE under a commercial pig farm condition in Korea. MATERIALS AND METHODS: Thirty two-day-old 672 piglets were randomly allocated into vaccinated and control groups. All piglets in the vaccinated group were inoculated with a commercial attenuated L. intracellularis vaccine as following the manufacturer's instruction. Body weights of all pigs in both groups were measured on the vaccination day and 6, 14, and 20 weeks post vaccination and an average daily weight gain (ADWG) was calculated. Health status was observed biweekly during the whole trial. RESULTS: The vaccinated group showed significantly higher body weight (p<0.05) and ADWG (p<0.05) than those of the control group. The vaccinated group had significantly reduced impairments in activity, growth, defecation frequency, and stool hardness (p<0.05). Additional health benefits and improved weight gain by the vaccination produced a 4.2:1 return of investment, and the higher gross margin was $4.80 per pig. CONCLUSION: Our finding suggests that the L. intracellularis vaccine program has effects on the substantial health and economic benefits in the Korean swine industry.

ABSTRACT

An outbreak of Lawsonia intracellularis infection in rabbits, which occurred in 1988 in Rio de Janeiro state, Brazil, is reported. The disease had an acute course (24-48 hours) with clinical signs characterized by brownish or green diarrhea and dehydration. Occasionally, the animals died one day after the onset of diarrhea, without showing any other clinical signs. At necropsy, the ileum was prominent, firm and had a thickened wall; it was dilated in the caudal direction and had a somewhat reticulated appearance, perceptible through the serosa. The thickened mucous membrane had finely corrugated aspect and a shiny surface. The ileocecal valve and surrounding areas were slightly edematous and irregular. The Peyer's patches were sometimes more evident. There was moderate enlargement of the mesenteric lymph nodes. The histological examination revealed different degrees of hyperplasia of the epithelial cells of intestinal crypts consisting of poorly differentiated, hyperchromatic cells with high mitotic index, arranged in a pseudostratified layer which, in some cases, reached the apical portions of the villi. The inflammatory infiltrate between the hyperplastic epithelial cells was composed of lymphocytes, plasma cells, macrophages, some eosinophils and globular leukocytes. Silver impregnation revealed large numbers of bacteria with morphology of the genus Lawsonia in the apical pole of cryptal enterocytes. These bacteria reacted positively to a Lawsonia intracellularis polyclonal antibody by the avidin-biotin immunohistochemistry method.


Descreve-se um surto de infecção por Lawsonia intracellularis em coelhos em Mendes, Estado do Rio de Janeiro. A doença manifestou-se, de forma aguda (24-48 horas), com sintomatologia caracterizada por diarréia marrom ou esverdeada, e desidratação. Ocasionalmente, os animais morriam um dia após o início da diarréia, sem apresentar outros sintomas. À necropsia verificou-se íleo proeminente, firme, com parede muito espessada, progressivamente dilatado no sentido caudal e com aspecto algo reticulado perceptível através da serosa. A mucosa espessada tinha aspecto finamente corrugado e superfície brilhante. A válvula íleo-cecal e imediações do ceco encontravam-se um pouco edemaciadas e irregulares. Por vezes, as placas de Peyer estavam maisevidentes. Observou-se também moderado aumento de volume dos linfonodos mesentéricos. O exame histológico revelou diferentes graus de hiperplasia das células epiteliais das criptas intestinais (células pouco diferenciadas, hipercromáticas, arranjadas de forma pseudo-estratificada e com alto índice mitótico) que, em parte dos casos, atingia a porção apical das vilosidades. A infiltração inflamatória, entre as células epiteliais hiperplásicas, era composta por linfócitos, plasmócitos, macrófagos, alguns eosinófilos e leucócitos globulóides. A impregnação por prata revelou grande número de bactérias com morfologia compatível com às do gênero Lawsonia no pólo apical dos enterócitos das criptas. ABSTRACT

Se realizó un estudio longitudinal para determinar la frecuencia e incidencia de Lawsonia intracellularis en cerdos de engorde en una granja de múltiples sitios y otra de un sitio en Yucatán, México. Se utilizaron 59 y 58 lechones negativos a L. intracellularis de las granjas de múltiples sitios y un sitio, respectivamente, los cuales se monitorearon cada 14 días de la semana, 1 a la 21 de edad. La bacteria en heces se detectó utilizando la prueba de la reacción en cadena de la polimerasa. Todos los cerdos de ambas granjas eliminaron L. intracellularis al menos una vez hasta la semana 11 de edad, encontrándose el mayor número de animales positivos a la semana 7. La incidencia acumulada fue del 100 por ciento en ambas granjas y la tasa de incidencia en las granjas de un sitio y múltiples sitios fueron 0,31 y 0,26 cerdos por cada 14 días a riesgo, respectivamente. Los resultados de este estudio proporcionan información para la implementación de esquemas de prevención control y erradicación de L. intracellularis.


A longitudinal study was carried out to determine the frequency and incidence of Lawsonia intracellularis in fattening pigs in a one-site farm and a multi-site farm in Yucatan, Mexico. Fifty-nine and 58 pigs negative to L. intracellularis from the one-site farm and the multi-site farm were used, respectively. Pigs were monitored every 14 days from weeks, one to 21 of age. The bacterium in faeces was detected by PCR test. All pigs from both farms shed L. intracellularis at least once by week 11 of age; the greatest number of positive pigs were detected at week 7. The cumulative incidence was 100% in both farms and the incidence density rates in the one-site farm and the multi-site farm were 0.31 and 0.26 pigs per each 14-days, respectively. The result of this study gives some information for the implementation of schemes of prevention, control and eradication of L. intracellularis.

ABSTRACT

Enteropatia proliferativa (EP), causada pela Lawsonia intracellularis, tem sido descrita em eqüinos jovens. A maioria dos relatos de EP em eqüinos é proveniente da América do Norte. Não existe ainda relato desta enfermidade em eqüinos na América Latina, apesar de a distribuição mundial da enfermidade em suínos. Portanto, é bastante provável que a EP esteja sendo negligenciada no diagnóstico de diarréias em potros desmamados. Esta revisão enfoca aspectos gerais sobre a infecção, abrangendo desde a etiologia, a epidemiologia, a patogenia, os sinais clínicos, as lesões anátomo e histopatológicas, o diagnóstico e o tratamento, alertando sobre a potencial importância de L. intracellularis como possível agente causal de diarréia em potros desmamados.


Proliferative enteropathy (PE), which is caused by Lawsonia intracellularis, has been recently described in young horses. The majority of horse PE cases have been reported in North America. Despite the fact that PE in swine has a worldwide distribution, there has not been any report of this disease in horse in Latin America yet. Therefore, it is very likely that L. intracellularis has been neglected on diagnosis of diarrhea in weanling foals. This review highlights general aspects about the infection, including etiology, epidemiology, pathogenesis, clinical signs, gross and histological lesions, diagnosis and treatment, and warns about the potential importance of L. intracellularis as possible causative agent of diarrhea in weanling foals.

Descrevem-se surtos de Enteropatia Proliferativa Hemorrágica causados por Lawsonia intracellularis em suínos no Estado do Rio de Janeiro. A sintomatologia caracterizou-se por diarréia sanguinolenta com evolução superaguda. À necropsia verificaram-se íleo de aspecto reticulado, com mucosa moderadamente espessada e grande quantidade de sangue parcialmente coagulado, além de marcada palidez de órgãos e carcaça. O exame histológico revelou proliferação hiperplásica das células epiteliais das criptas de Lieberkühn, por vezes, associada a alterações inflamatórias e necróticas. A imunohistoquímica demonstrou presença da bactéria, em grande quantidade, dentro do citoplasma das células epiteliais das criptas. L. intracellularis também foi visualizada pela ultramicroscopia. Os surtos ocorreram em 1987 e essa é a primeira descrição da enfermidade no Estado do Rio de Janeiro.


Outbreaks of infection of swine with Lawsonia intracelullaris in the state of Rio de Janeiro are described. The symptomathology was characterized by bloody diarrhea with peracute course. Postmortem findings were pale organs and tissues, reticulate aspect of the ileum with its mucosa moderately thickened nad contained large amounts of partially coagulated blood. Histopathological examination revealed hyperplastic epithelial cell proliferation of the Lieberkühn crypts, sometimes associated with inflammatory and necrotic alterations. Immunohistochemistry demonstrated the presence of large amounts of bacteria in the cytoplasm of the cryptal epithelial cells. L. intracellularis was also seen by ultramicroscopic examination. These outbreaks occurred in 1987 and this is the first report of the disease in the state of Rio de Janeiro.

ABSTRACT

Estudou-se a eficácia do diagnóstico macroscópico de lesões da enteropatia proliferativa suína (EPS) usando-se fragmentos de íleo de 663 suínos, coletados em abatedouros localizados em três municípios do Rio Grande do Sul. As amostras foram processadas por métodos histológicos rotineiros e coradas por uma técnica desenvolvida pela combinacão das coloracões Warthin-Starry, alcian blue e hematoxilina-eosina para deteccão simultânea de Lawsonia intracellularis e lesões associadas com EPS. Lâminas suspeitas de EPS foram submetidas à técnica de imunoistoquímica, utilizando anticorpo policlonal anti-Lawsonia intracellularis na diluicão de 1:15.000 pelo método avidina-biotina. A coloracão combinada detectou 11 casos positivos, e a imunoistoquímica, nove casos adicionais. Entre as 643 amostras consideradas negativas, 12 apresentaram desaparecimento de células caliciformes e proliferacão adenomatosa características de EPS, mas ausência de bactérias intracelulares. A eficiência do exame macroscópico para diagnóstico de EPS foi medida pela associacão entre os resultados das avaliacões macroscópicas e histológicas realizadas em 219 amostras. Embora 51 delas tenham sido consideradas macroscopicamente positivas, apenas quatro foram confirmadas pela presenca de bactérias intracelulares associadas com lesões características de EPS. Não se observou associacão entre as alteracões macroscópicas e histológicas de EPS.

ABSTRACT

Avaliou-se a distribuição de Lawsonia intracellularis em fezes de suínos, coletadas diretamente no reto, durante a fase de crescimento/terminação, sob condições de campo, e determinou-se o efeito da infecção L. intracellularis no ganho de peso. O estudo epidemiológico longitudinal foi desenvolvido em uma granja de múltiplos sítios em Yucatan, México. A granja possuía três sítios geograficamente isolados com o sistema "todos dentro todos fora". Uma amostra de 56 suínos negativos para a bactéria foi calculada utilizando prevalência esperada de 5 por cento, nível de confiança de 95 por cento e população de 2000 suínos terminados. Os suínos foram amostrados 12 vezes desde o nascimento até a 23ª semana de idade. A detecção de DNA bacteriano nas fezes pela técnica de reação em cadeia pela polimerase em qualquer coleta representou um caso. Dos 56 suínos monitorados, 13 eliminaram L. intracellularis nas fezes e somente um resultou positivo duas vezes. A eliminação de L. intracellularis foi detectada desde a terceira até a 19ª semana de idade, observando-se maior número de animais positivos na 11ª semana. A incidência acumulada e a taxa de incidência foram 23,2 por cento e 1,21 suínos por 100 animais/semana em risco, respectivamente. As médias do ganho de peso por dia foram 0,742±0,06kg e 0,754±0,07kg para os suínos negativos e positivos, respectivamente (P>0,05). A eliminação da bactéria ocorreu durante todo o período de estudo. Pelo menos neste estudo, L. intracellularis não pode ser utilizado para predizer o efeito da bactéria sobre o ganho de peso.

ABSTRACT

A multiplex PCR assay was developed for the simultaneous detection of the etiologic agents associated with porcine proliferative enteropathies (PPE), swine dysentery (SD)and porcine salmonellosis (PS)in a single reaction using DNA from swine intestinal samples. Single and multiplex PCR amplification of DNA from Lawsonia intracellularis, Salmonella typhimurium and Brachyspira hyodysenteriae with each primer set produced fragments of the predicted size without any nonspecific amplification, 210-bp, 298-bp and 403-bp bands, respectively. The single PCR assay could detect as little as 100 pg of purified DNA of S. typhimurium and L. intracellularis, and 50 pg of B.hyodysenteriae, respectively. However, multiplex PCR turned out to be 10 times lower sensitivity with S. typhimurium compared with single PCR. With 23 swine intestinal specimens suspected of having PPE, SD and/or PS, the multiplex PCR assay showed identical results with conventional methods except one. In conclusion, this multiplex PCR is a feasible alternative to standard diagnostic methods for detection of L. intracellularis, B. hyodysenteriae and Salmonella spp. from swine intestinal specimens.

ABSTRACT

Proliferative enteropathy was reproduced in IFN-gamma receptor knockout (IFN-gamma R-) mice by experimental infection with Lawsonia intracellularis (L. intracellularis). The cecum and the colon of the infected mice were evidently enlarged 2 weeks post infection. The presence of L. intracellularis was identified in the stool and the cecum of the mice after infection. However, high levels of IFN-gamma were detected in the sera of the infected mice 2 weeks PI. These data indicated that the IFN-gamma produced in the infected mice should have been utilized by it's receptor to elicit protective immune responses against L. intracellularis infections.

ABSTRACT

The prevalence of Lawsonia intracellularis, Brachyspira hyodysenteriae and Salmonella spp. were investigated by multiplex PCR using fecal samples of pigs with diarrhea or a history of diarrhea. The overall herd prevalence of L. intracellularis, B. hyodysenteriae and Salmonella spp. were 46.5%, 37.2% and 51.1%, respectively. Also, the prevalence of L. intracellularis, B. hyodysenteriae and Salmonella spp. among all sampled pigs were 19.9%, 10.8% and 17.7%, respectively. Seventeen of 43 herds were positive with 2 enteric organisms, and 2 herds were positive with L. intracellularis, B. hyodysenteriae and Salmonella spp. simultaneously. It was notable that 11 of 12 herds with more than 2, 000 pigs were affected with Salmonella spp., and that only 2 of 12 the herds were affected with B. hyodysenteriae. This study suggested that herds positive for L. intracellularis, B. hyodysenteriae and Salmonella spp. were distributed throughout Korea, although the relationship among other pathogens such as viral or parasitic ones and/or with metabolic disorders was not determined.

En el período comprendido entre julio y octubre del año 1997 se evaluaron 360 intestinos de cerdos sacrificados en el Matadero Municipal de Medellín, Colombia, con el objetivo de establecer la prevalencia de la Enteropatía Proliferativa Porcina (EPP) en el departamento de Antioquia, caracterizar las lesiones anatomopatológicas asociadas y determinar la presencia de su agente etiológico Lawsonia intracellularis. El cálculo de la muestra se efectuó de una población tabular infinita con un límite de confianza del 95 por ciento y una seguridad de ±5 por ciento. Las muestras obtenidas se sometieron a inspección macroscópica y evaluación histopatológica utilizando las coloraciones de Hematoxilina-Eosina y Warthin-Starry. El análisis de los resultados se efectuó mediante estadística descriptiva y la prueba de independencia y contingencia (chi-cuadrado). La prevalencia de EPP fue del 87.22 or ciento, determinada por la presencia de las lesiones características de la enfermedad asociadas a su agente etiológico L. intracellularis. Las formas lesionales más frecuentes fueron: Adenomatosis Intestinal en un 74.72 por ciento e Ileítis Regional en un 20.55 por ciento. Este artículo discute los hallazgos epidemiológicos y anatomopatológicos de EPP en el departamento de Antioquia y se constituye en el primer reporte del hallazgo de una bacteria como L. intracellularis diagnosticada por un método directo en Colombia.

ABSTRACT

A enterite proliferativa suína, a espiroquetose colônica, a disenteria suína e a salmonelose são as doencas entéricas mais freqüentes em suínos das fases de crescimento e terminacão no Brasil. O diagnóstico destas doencas através de técnicas bacteriológicas tradicionais é difícil, lento e no caso da enterite proliferativa suína, impossível. A deteccão destes agentes através da reacão de polimerase em cadeia é altamente sensível e específica e está se tornando uma ferramenta muito útil no diagnóstico em Medicina Veterinária. No presente estudo foram testadas duas técnicas de PCR sob a forma de "multiplex"para deteccão e identificacão simultânea dos agentes bacterianos envolvidos na enterite proliferativa suína, na espiroquetose colônica, na disenteria suína e na salmonelose em amostras de fezes diarreicas. O DNA obtido de culturas puras de cada agente sozinho ou misturado em diferentes combinacões e concentracões foram amplificados utilizando a Multiplex PCR específica para Lawsonia intracellularis e Salmonella, ou Brachispyra pilosicoli e Brachispyra hyodysenteriae. Após a padronizacão, a M-PCR foi aplicada na deteccão dos quatro agentes em 541 amostras de fezes diarreicas de suínos provenientes de diferentes Estados do Brasil. O agente mais freqüente foi Lawsonia intracellularis (13 per center), seguido pela Salmonella spp. (4,8 per center), B. hyodysenteriae (1,4 oer center), B. pilosicoli (1 per center) e suas diferentes associacões. Os resultados obtidos indicam que as duas reacões testadas permitem a deteccão destes quatro importantes patógenos entéricos de maneira rápida e específica.
