RESUMEN

Rauwolfia radix was produced both via callus and directly, using stem as an explant. MS medium containing 1 mg/1 NAA and 0.5 mg/1 Kn with 1% CH was required for proliferation of callus and initiation of root projections [visible root primordial]. Continuous [24 h] illumination was required for development of root projections into true roots. Roots formed via callus or direct on stem explant were distinguishable from each other. WRC medium was employed for root propagation, with addition of 0.8 mg/l NAA and 10 mg/l biotin

In vitro Rauwolfia roots were differentiated via regenerated plant's leaf callus cultures, grown on WRC medium containing 10% CM and 0.1 mg/l each of NAA and BAP. RESUMEN

Callus cultures were initiated from redical, leaf, petiole, stem and root explants of H. niger. MS medium with kinetin and 2,4-D at a concentration of 0.1 mg/1 was found suitable for good amount of callus formation. Callus was propagated in undifferentiated form, for more than a year with the addition of CH, CM and SDC to the culture medium. The presence of alkaloids in cultures was observed dependent on the colour and origin of the callus
